摘  要:目的研究TGFN—Smad3信号通路在胃癌条件培养基(CM)诱导网膜脂肪干细胞(O—ASCs)分化为癌相关成纤维细胞(CAFs)中的作用。方法通过诱导分化成骨、成脂及流式细胞鉴定O—ASCs,将O—ASCs分为对照组、SGC7901-CM试验组、SGC7901-CM+SB431542处理组,12h后收集细胞。通过RT—PCR和Western—blot检测各组细胞CAFs标志物α—SMA、FSP-1的表达水平。通过Western—blot检测Smad信号通路Smad3、P—Smad3的表达水平。结果鉴定原代培养细胞为O—ASCs。与对照组相比,SGC7901-CM组CAFs特异性标志物仪-SMA、FSP-1及Smad信号通路蛋白P—Smad3均增加明显(P〈0.05),Smad3表达未见明显变化(P〉0.05)。与SGC7901-CM组相比,SGC7901-CM+SB431542组仪-SMA,FSP-1及P—Smad3蛋白表达均受到明显抑制(P〈0.05),Smad3表达未见明显变化(P〉0.05)。结论胃癌条件培养基具有诱导O—ASCs向CAFs分化的作用,TGFβ1-Smad3信号通路在该过程中发挥了重要作用。Objective To investigate effects of TGFβ1 - Smad3 signal pathway on the differentiation of omental - adipose stromal ceils( O- ASCs)towards carcinoma- associated fibroblasts (CAFs)induced by gastric cancer condi- tioned medium(CM). Methods O - ASCs were identified by examining their ability to differentiate into osteogenic and adipogenic lineages and flow cytometry. O - ASCs were divided into control group, SGC7901 - CM group and SGC7901 -CM + SB431542 group. Cells were collected after 12 hours. RT- PCR and western- blot were utilized to evaluate the expression of α - SMA and FSP - 1. Western-blot was utilized to evaluate the expression of Smad3 and p - Smad3. Results Primary ceils were verified as O - ASCs. The expression of α - SMA, FSP - 1 and p - Smad3 in- creased( P 〈 0.05 ) and the expression of Smad3 didnt change significantly ( P 〉 0.05 ) in SGC7901 - CM group than those in control group. The expression of α - SMA, FSP - 1 and p - Smad3 decreased ( P 〈 0.05 ) and the expression of Smad3 didnt change significantly( P 〉0.05 )in SGC7901 -CM + SB431542 group than those in SGC7901 -CM group ( P 〈 0.05). Conclusion O - ASCs can be induced to CAFs exposed to gastric cancer conditioned medium. TGFI31 - Smad3 sienal uathwav olavs an imoortant role in the differentiation of O -ASCs towards CAFs.
