摘 要:目的:研究三氧化二砷(As2_O_3)联合佛波酯(PMA)对急性早幼粒细胞白血病细胞系Kasumi-1增殖的影响。方法:培养急性早幼粒细胞白血病细胞系Kasumi-1并随机分为对照组(不含药物和血清的RPMI1640培养基处理)、As2_O_3组(含有20μmol/L As2_O_3的无血清RPMI1640培养基处理)、PMA组(含有160nmol/L PMA的无血清RPMI1640培养基处理)、As2_O_3+PMA组(含有20μmol/L As2_O_3和160nmol/LPMA的无血清RPMI1640培养基处理)。处理后测定细胞的增殖活力、细胞周期的比例以及相关基因的蛋白表达量。结果:干预后12、24、48h时,As2_O_3组、PMA组、As2_O_3+PMA组的细胞增殖活力均显著低于对照组,As2_O_3+PMA组的细胞增殖活力均显著As2_O_3组、PMA组;干预后48h时,As2_O_3组、PMA组、As2_O_3+PMA组中G1期、S期的比例以及CDK1、CyclinB1的表达量均显著低于对照组,G2期的比例以及Bax、Caspase-3、Caspase-9、p-Chk1、p-Cdc25C9的表达量均显著高于对照组;As2_O_3+PMA组中G1期、S期的比例以及CDK1、CyclinB1的表达量均显著低于As2_O_3组、PMA组,G2期的比例以及Bax、Caspase-3、Caspase-9、p-Chk1、p-Cdc25C的表达量均显著高于As2_O_3组、PMA组。结论:As2_O_3联合PMA能够通过诱导细胞凋亡、阻碍细胞周期的方式抑制急性早幼粒细胞白血病细胞系Kasumi-1的增殖。Objective:To study the effect of arsenic trioxide(As2O3)combined with phorbol 12-myristate 13-acetatefor(PMA)on the proliferation of acute promyelocytic leukemia cell line Kasumi-1.Methods:Acute promyelocytic leukemia cell lines Kasumi-1were cultured and randomly divided into control group(treated with the RPMI1640 medium without drugs or serum),As2O3group(treated with serum-free RPMI1640 medium containing 20μmol/L As2O3),PMA group(treated with serum-free RPMI1640 medium containing 160nmol/L PMA)and As2O3+ PMA group(treated with serum-free RPMI1640 medium containing 20μmol/L As2O3 and 160nmol/L PMA).After treatment,the cell proliferation activity,cell cycle ratio and the protein expression of related genes were measured.Results:At 12 h,24hand 48 hafter treatment,the cell proliferation activity of As2O3 group,PMA group and As2O3+PMA group were significantly lower than that of control group,and the cell proliferation activity of As2O3+PMA group was significantly lower than that of As2O3 group and PMA group;48hafter treatment,the G1 phase and S phase ratio as well as CDK1 and CyclinB1expression of As2O3 group,PMA group and As2O3+PMA group were significantly lower than those of control group while the G2 phase ratio as well as Bax,Caspase-3,Caspase-9,p-Chk1 and p-Cdc25C9 expression were significantly higher than those of control group;the G1 phase and S phase ratio as well as CDK1 and CyclinB1expression of As2O3+PMA group were significantly lower than those of As2O3 group and PMA group while the G2 phase ratio as well as Bax,Caspase-3,Caspase-9,p-Chk1 and p-Cdc25C9 expression was significantly higher than those of As2O3 group and PMA group.Conclusions:As2O3combined with PMA can inhibit the proliferation of acute promyelocytic leukemia cell line Kasumi-1by inducing apoptosis and blocking cell cycle.
