摘 要:目的:研究肾癌患者血清和组织中miR-21,-122,-222的含量并探究其潜在靶基因。方法:选择在本院诊断为肾癌并接受手术切除的54例患者及同期体检的70例健康志愿者作为本研究的受试者,分别作为肾癌组和对照组,采集血清标本及肾癌组织、癌旁组织。检测血清、肾癌组织、癌旁组织中miR-21、-122、-222的表达量以及肾癌组织、癌旁组织中抑癌基因、侵袭抑制基因的表达量。结果:肾癌组血清中miR-21,-122,-222的表达量显著高于对照组;肾癌组织中miR-21,-122,-222的表达量显著高于癌旁组织,PDCD4、BTG1、NPRL2、Bax、TIMP1、TIMP2、E-cadherin的表达量低于癌旁组织。miR-21,-122,-222的表达量与PDCD4、BTG1、NPRL2、Bax、TIMP1、TIMP2、E-cadherin的表达量呈负相关。结论:肾癌患者血清和组织中高表达的miR-21,-122,-222能够抑制多种抑癌基因、侵袭抑制基因的表达,进而促进肾癌细胞的增殖、侵袭。Objective:To study the miR-21,-122and-222 contents in serum and tissues of patients with renal cell carcinoma and explore their potential target genes.Methods:A total of 54 cases of patients diagnosed as renal cell carcinoma and received surgical resection in Shenzhen Second People's Hospital during May 2014-December 2016 and 70cases of healthy volunteers received physical examination during the same period were selected as renal carcinoma group and control group.Then serum samples,renal carcinoma tissues and paracancerous tissues were collected.The expression levels of miR-21,-122and-222 in serum,renal carcinoma tissues,paracancerous tissues and and the expression of tumor suppressor gene and invasion suppressor gene in renal carcinoma tissues and adjacent tissues were detected.Results:Serum miR-21,-122,-222 expression of renal cell carcinoma patients were significantly higher than the control group;miR-21,-122,-222 expression in renal cell carcinoma tissues were significantly higher than paracancerous tissues,PDCD4,BTG1,NPRL2,Bax,TIMP1,TIMP2,E-cadherin expression were lower than paracancerous tissues.The expression of miR-21,-122,-222 were negatively correlated with the expression of PDCD4,BTG1,NPRL2,Bax,TIMP1,TIMP2,E-cadherin.Conclusion:High expression of miR-21,-122and-222 in serum and tissues of renal cell carcinomapatients can inhibit the expression of a variety of tumor suppressor genes and invasion suppressor genes,thereby promoting the proliferation and invasion of renal carcinoma cells.
