摘 要:采用单因素试验结合正交试验的方法对杠板归多糖提取工艺进行优选,并对其羟自由基清除作用、超氧阴离子清除作用和还原能力进行研究.结果表明,最佳提取工艺为:酶浓度5 mg/mL、温度50℃、水料比30∶1(mL/g)、时间120 min.在此工艺条件下,杠板归多糖的提取率可达3.68%.杠板归多糖对羟自由基的最大清除率为78.56%,IC_(50)=0.442 mg/mL,对超氧阴离子的最大清除率为89.81%,IC_(50)=0.297 mg/mL,杠板归多糖还具有一定的还原能力.The extraction technology of Polygonum perfoliatum L. polysaccharide with a single-factor test and orthogonal was optimized. The hydroxyl free radical scavenging effect, superoxide anion scavenging effect and reducing power of Polygonum pe(oliatum L. polysaccharide were studied. The experimental result indicates that the optimal extraction condition is when cellulase concentration is 5 mg/mL, extraction temperature is 50℃, water material ratio is 30:1 (mL/g), extraction time is 120 rain. Under these conditions, the yield of Polygonum perfoliatum L. polysaccharide is 3.68%. The maximum scavenging rate of polysaccharide to hydroxyl radical is 78.56%, IC50 = 0.442 mg/mL. The maximum scavenging rate of polysaccharide to superoxide anion is 89.81%, IC50 = 0.297 mg/mL. Polygonum perfoliatum L. polysaccharide also has a certain reduction force.
