摘 要:目的通过肺组织病理学观察及渗出液成分检测,探讨硫化氢(H_2S)对实验性高原肺水肿大鼠呼吸膜通透性的影响。方法健康成年雄性Wistar大鼠随机分为四组:常氧对照组、低氧对照组、试剂(PPG)常氧组、试剂(PPG)低氧组。用面罩低氧方式构建大鼠实验性高原肺水肿模型。用腹腔注射PPG抑制内源性H_2S的产生。用ELISA法检测肺组织中EB含量和支气管肺泡灌洗液(BALF)中TP含量,光镜下观察肺组织的病理学改变。结果 PPG常氧组中BALF的TP含量明显高于常氧对照组(P<0.05),PPG低氧组肺组织EB含量和BALF中TP含量明显高于低氧对照组和PPG常氧组(P<0.05);镜下观察到PPG低氧组肺组织病理损伤最为严重,PPG常氧组较常氧对照组严重。结论降低内源性H_2S可增加实验性高原肺水肿大鼠肺呼吸膜通透性,导致蛋白质渗出增多;体内H_2S的减少会加重低氧对肺呼吸膜的损伤。Objective To investigate the effect of hydrogen sulfide on pulmonary respiratory membrane permeability in rats with experimental high altitude pulmonary edema by detecting lung histopathology and the composition of pulmonary effusion fluid.Methods Healthy adult male Wistar rats were randomly divided into 4 groups:normoxia group,hypoxia group,PPG normoxia group and PPG hypoxia group.The models of rat with experimental high alti- tude pulmonary edema were established by hypoxie mask ,the concentration of H2S in vivo was reduced by intraper- itoneal injection of PPG.The contents of total protein (TP)in broncho alveolar lavage fluid (BALF)and the Evans blue(EB) in lung tissue were measured by ELISA.The pathological changes of lung tissue were observed under light microscope.Results The content of TP in BALF in PPG normoxia group was significantly higher than those in nor- moxia group(P〈0.05).The content of TP in BALF and the EB in lung tissue of PPG hypoxia group were significantly higher than those in hypoxia group and PPG normoxia group(P〈0.05).The pathological lesion in PPG nor- moxia group was more serious than those in normoxia group, and the pathological lesion in PPG hypoxia group was the most serious.Conclusion The decrease of H^S concentration could increase the pulmonary respiratory membrane permeability and lead to the increase of protein excretion.Meanwhile the reduction of H2S could aggravate the damage of pulmonary respiratory membrane permeability in hypoxia.
