摘  要:目的分析人宫颈癌HeLa细胞核内M-CSF增高对化疗药物的影响。方法体外培养HeLa细胞、HeLa-M细胞以及转染M-CSF空载体HeLa-C细胞。分别使用紫杉醇和顺铂(浓度分别为0.3、3、30μg/m L)作用48 h,采用MTT方法比较细胞的相对活性,Westen blot分析转染M-CSF对抗凋亡分子Bcl-2表达的影响。结果当两种化疗药物的浓度为3、30μg/m L时,与HeLa-C细胞和HeLa细胞相比,HeLa-M细胞活性明显增高,差异有统计学意义(P<0.05);当浓度为0.3μg/m L时三种细胞活性无明显差异。Westen blot结果显示,转染M-CSF后能够使细胞中的抗凋亡蛋白Bcl-2表达水平明显升高。结论人宫颈癌HeLa细胞核中的M-CSF增高可以使细胞的药抗性提高,从而抑制HeLa细胞的凋亡。Objective To analyze the effect of elevated M-CSF in nuclei on the chemotherapeutic drugs of human cervical cancer HeLa cells. Methods HeLa cells, HeLa-M cells and HeLa-C transfected with M-CSF empty vector were cultured in vitro. The paclitaxel and cisplatin(concentrations were 0.3, 3, 30 μg/mL) were used for 48 h, respectively. The relative viability of the cells was compared by MTT method. The effect of transfection with M-CSF on the expression of Bcl-2 was detected by Western blot analysis. The effect of paclitaxel and cisplatin(concentration 0.3, 3, 30 μg/mL) on the expression of anti-apoptotic molecule Bcl-2 was detected by MTT method. Results The viability of HeLa-M cells was significantly higher than that of HeLa-C cells and HeLa cells when the concentration of the two chemotherapeutic drugs was 3, 30 μg/mL, and the difference was statistically significant(P0.05). There was no significant difference in cell viability between the three kinds of cells when the concentration was 0.3 μg/mL. The results of Western blot showed that the expression of anti-apoptotic protein Bcl-2 was significantly increased after transfection with M-CSF. Conclusion The increase of M-CSF in the nucleus of human cervical cancer HeLa cells can improve the drug resistance of the cells and inhibit the apoptosis of HeLa cells.
