摘  要:目的探讨WAVE3和上皮型钙黏蛋白(E-cadherin)在结直肠癌中的表达及与结直肠癌肝转移的相关性。方法将2008年1月至2013年6月内蒙古科技大学包头医学院第一附属医院收治的结直肠癌患者,按是否有肝转移分为非肝转移组(48例)和肝转移组(48例)。对照组:48例为上述病例中数字法抽取的癌旁>5 cm的正常黏膜组织。应用免疫组化法检测WAVE3和E-cadherin在结直肠癌及癌旁组织中的表达,分析两者在结直肠癌中的表达及与肝转移的相关性。结果 (1)WAVE3在结直肠癌不伴肝转移及结直肠癌伴肝转移患者的原发灶组织中的表达阳性率高于癌旁正常黏膜组织(P<0.05);WAVE3在结直肠癌伴肝转移患者原发灶组织中的表达阳性率低于结直肠癌不伴肝转移的表达(P<0.05);(2)E-cadherin在结直肠癌不伴肝转移及结直肠癌伴肝转移的患者的原发灶组织中的表达阳性率低于癌旁正常黏膜组织(P<0.05);E-cadherin在结直肠癌伴肝转移的表达阳性率低于结直肠癌不伴肝转移的表达(P<0.05);(3)Spearman相关分析显示,WAVE3和E-cadherin表达的阳性率无显著相关性(r=-0.199,P>0.05);(4)WAVE3的表达均与肿瘤的淋巴结转移和Dukes分期有关(P<0.05),E-cadherin的表达均与肿瘤的分化程度,淋巴结转移和Dukes分期有关(P<0.05)。结论 WAVE3和E-cadherin在结直肠癌肝转移患者结直肠癌组织中表达降低与结直肠癌侵袭、转移、预后相关,联合检测WAVE3和E-cadherin在结直肠癌中的表达,可能有助于预防结直肠癌肝转移的发生。Objective To detect the expression of WAVE3 and E-cadherin in colorectal carcinoma (CRC) and to investigate its correlation with colorectal carcinoma liver metastasis (CRLM). Methods A total of 48 CRC cases and 48 CRLM cases were selected randomly as research objects. Adjacent normal laryngeal mucosa ( 〉 5 cm) of 48 cases from the patients above were randomly selected as the control group. The expres- sions of WAVE3 and E-cadherin in resection specimens of colorectal cancer tissues and adjacent tissues were as- sayed by immunohistochemistry staining method. Correlations between of the expressions with liver metastasis were also analyzed. Results (1)The positive expression rates of WAVE3 protein in CRC and CRLM were sig- nificantly higher than that in adjacent normal mucosa tissues ( both P 〈 0. 05). The expression of WAVE3 in CRLM was significantly lower than that of CRC (P 〈 0. 05). (2)The positive expression rates of E-cadherin pro- tein in CRC and CRLM were significantly lower than that in adjacent normal mucosa tissues ( both P 〈 0. 05 ). The expression of E-cadherin in CRLM was significantly lower than that of CRC (P 〈 0. 05 ). (3)The positive ex-pression rates of WAVE3 and E-cadherin had no significant correlation in Spearman relative analysis (r = -0. 199, P 〉 0. 05). (4)The expression of WAVE3 was associated with lymph node metastasis and Dukes stage (both P 〈 0. 05 ). The expression of E-cadherin protein was correlated with the tumor differentiation degree, lymph node metastasis and Dukes staging ( all P 〈 0. 05 ). Conclusions Synchronously low expressions of WAVE3 and E-cadherin in colorectal carcinoma were associated with liver metastasis, which provide a new method for early prevention or early diagnosis of CRLM.
