摘 要:目的观察候选抑癌基因SASH1与磷酸化的细胞外信号调节激酶1/2(p-ERK1/2)在胃腺癌中的表达、意义及二者的相互关系。方法免疫组织化学法检测152例胃腺癌组织及30例癌旁组织SASH1和p-ERK的表达。分析SASH1、p-ERK与胃癌临床病理学特征的关系及SASH1与p-ERK蛋白表达的相关性。结果 SASH1在胃癌组织中的阳性率显著低于癌旁组织(P<0.01),在胃低分化腺癌中的阳性率显著低于胃高、中分化腺癌(P<0.01)。SASH1阳性率在Ⅲ-Ⅳ期胃癌患者、有淋巴结转移患者及有远端转移患者分别显著低于Ⅰ-Ⅱ期患者、无淋巴结转移患者及无远端转移患者(P<0.05,P<0.01;P<0.05)。p-ERK1/2在胃癌组织中的阳性率显著高于癌旁组织(P<0.01),在胃低分化腺癌中的阳性率显著高于胃高、中分化腺癌(P<0.01)。p-ERK1/2阳性率在Ⅲ-Ⅳ期胃癌患者、有淋巴结转移患者及有远端转移患者分别显著高于Ⅰ-Ⅱ期患者、无淋巴结转移患者及无远端转移患者(P值分别为P<0.05;P<0.05;P<0.01)。SASH1与p-ERK在胃腺癌中的表达呈显著负相关(P<0.05)。结论SASH1蛋白可能通过调控ERK1/2信号通路抑制胃癌进展。Purpose To investigate the expression and significance of SASH1, a candidate tumor suppressor gene, and phosphorylated extraeellular signal-regulated kinase (p-ERK1/2) in gastric adenocareinoma, and to explore the relationship between these proteins. Methods The expression of SASH1 and p-ERK was immunohistochemically detected in 152 eases of gastric adenocarcinoma and 30 eases of adjacent tissues. The relationship between SASH1, p-ERK and the clinicopathological characteristics of gastrie careinoma and the correlation between the expression of SASH1 and p-ERK were analyzed. Results The positive rate of SASH1 expression was significantly lower in gastric carcinoma than that in adjacent tissues ( P 〈 0. 05 ), and significantly lower in poorly differentiated gastrie adenoeareinoma than that in well- moderately differentiated gastric adenocarcinoma ( P 〈 0. 05 ). The positive rate of SASH1 expression was significantly lower in patients with III-IV phase gastric cancer, having lymph node metastasis and having distant metastasis than those having I- II phase gastric cancer, without lymph node metastasis and without distant metastasis, respectively (P 〈 0.05; P 〈 0. 01; P 〈 0. 05 ). The positive rate of p-ERK1/2 expression was significantly higher in gastric carcinoma compared with that in adjacent tissues ( P 〈 0. 01 ) , and significantly higher in poorly differentiated gastric adenoeareinoma than that in well- moderately differentiated gastric adenoeareinoma ( P 〈 0. 01 ). The positive rate of SASH1 expression was significantly higher in patients with III-IV phase gastric eaneer, having lymph node metastasis and having distant metastasis than those having I-II phase gastric cancer, without lymph node metastasis and without distant metastasis, respectively ( P 〈 0. 05 ; P 〈0. 05; P 〈 0. 01 ).
