摘  要:目的:研究马尾神经压迫损伤后细胞凋亡相关蛋白、氧化应激损伤相关蛋白的表达规律。方法:选择雄性本地家犬作为实验动物并随机分为压迫组、假手术组,通过水囊压迫的方法建立马尾神经压迫损伤模型。压迫后24、48、72h,分离压迫损伤的马尾神经并测定细胞凋亡相关蛋白、氧化应激损伤相关蛋白的含量。结果:压迫后24、48、72h,压迫组家犬马尾神经中Bcl-2的蛋白表达量显著低于假手术组,Bax、Cyt-C、Caspase-3、Caspase-9、Beclin-1、LC3-II的蛋白表达量以及活性氮(RNS)、活性氧(ROS)、丙二醛(MDA)、晚期氧化蛋白产物(AOPP)、8-羟基脱氧鸟苷酸(8-OHdG)的含量均显著高于假手术组。结论:马尾神经压迫损伤会造成线粒体细胞凋亡、细胞自噬、氧化应激的过度激活。Objective: To study the expression pattern of apoptosis related proteins and oxidative stress related proteins after cauda equina compression injury.Methods: Male domestic dogs were selected as experimental animals and randomly divided into compression group and sham operation group.Cauda equina compression injury model was established by water sac compression.Of 24 hours, 48 hours and 72 hours after compression, the cauda equina nerve was isolated and the contents of apoptosis related proteins and oxidative stress related proteins were measured.Results: Of 24 hours, 48 hours, 72 hours after the compression, the protein expression of Bcl-2 in dog cauda equina of compression group was significantly lower than the sham group, the protein expression of Bax, Cyt-C, Caspase-3, Caspase-9, Beclin-1, LC3-II and the content of RNS, ROS, MDA, AOPP, 8-OHdG were significantly higher than those of sham operation group.Conclusions: the injury of cauda equina nerve can cause excessive activation of mitochondria apoptosis, autophagy and oxidative stress.
