摘  要:目的:探讨膀胱癌组织中MCP-1、VEGF、CUGBP1表达相关性及其与癌细胞生长的关系。方法:收集在本院接受膀胱癌根治术治疗的原发性膀胱癌患者80例,取膀胱癌组织及癌旁组织,采用western-blot法测定其中MCP-1、VEGF、CUGBP1的蛋白表达量,采用荧光定量PCR法测定其中增殖、凋亡基因的mRNA表达量。经Pearson检验评估MCP-1、VEGF、CUGBP1的相关关系,以及MCP-1、VEGF、CUGBP1蛋白表达量与增殖、凋亡基因mRNA表达量的相关关系。结果:膀胱癌组织中MCP-1、VEGF、CUGBP1的蛋白表达量显著高于癌旁组织,且三者蛋白表达量间互呈正相关关系(P<0.05)。膀胱癌组织中增殖基因KPNA2、MRPS5、YAP的mRNA表达量高于癌旁组织,NDPK-A、Sox1的mRNA表达量低于癌旁组织,凋亡基因Bcl-2、Rce1、cIAP-1的mRNA表达量高于癌旁组织,Bax、STAG2、RUNX3的mRNA表达量低于癌旁组织(P<0.05)。膀胱癌组织中MCP-1、VEGF、CUGBP1的蛋白表达量与增殖基因KPNA2、MRPS5、YAP的mRNA表达量呈正相关,与NDPK-A、Sox1的mRNA表达量呈负相关,与凋亡基因Bcl-2、Rce1、cIAP-1的mRNA表达量呈正相关,与Bax、STAG2、RUNX3的mRNA表达量呈负相关(P<0.05)。结论:膀胱癌组织中MCP-1、VEGF、CUGBP1互相促进表达,且共同作用于肿瘤细胞的增殖、凋亡基因,加速肿瘤细胞增殖并抑制其凋亡。Objective: To study the MCP-1, VEGF and CUGBP1 expression correlation in bladder cancer tissue and their relationship with the cancer cell growth.Methods: A total of 80 patients with primary bladder cancer who accepted radical operation for bladder cancer in our hospital between July 2012 and July 2016 were selected, the bladder cancer tissue and tissue adjacent to carcinoma were collected, western-blot method was used to determine the MCP-1, VEGF and CUGBP1 protein expression and fluorescent quantitative PCR method was used to determine proliferation and apoptosis gene mRNA expression.Pearson test was used to assess the correlation among MCP-1, VEGF and CUGBP1 as well as the correlation of MCP-1, VEGF and CUGBP1 protein expression with proliferation and apoptosis gene mRNA expression.Results: MCP-1, VEGF and CUGBP1 protein expression in bladder cancer tissue were significantly higher than those in tissue adjacent to carcinoma, and the protein expression of the three were positively correlated (P〈0.05).Proliferation genes KPNA2, MRPS5 and YAP mRNA expression in bladder cancer tissue were higher than those in tissue adjacent to carcinoma while NDPK-A and Sox1 mRNA expression were lower than those in tissue adjacent to carcinoma, and apoptosis genes Bcl-2, Rce1 and cIAP-1 mRNA expression were higher than those in tissue adjacent to carcinoma while Bax, STAG2 and RUNX3 mRNA expression were lower than those in tissue adjacent to carcinoma (P〈0.05).MCP-1, VEGF and CUGBP1 protein expression in bladder cancer tissue were positively correlated with proliferation genes KPNA2, MRPS5 and YAP mRNA expression while negatively correlated with NDPK-A and Sox1 mRNA expression, and were positively correlated with apoptosis genes Bcl-2, Rce1 and cIAP-1 mRNA expression while negatively correlated with Bax, STAG2 and RUNX3 mRNA expression (P〈0.05).Conclusions: MCP-1, VEGF and CUGBP1 expression in bladder cancer tissue promote each other, act on tumor cell proliferation and apoptosis genes together, accelera
