子宫内膜癌组织辣椒素受体TRPV1表达临床意义探讨

摘　　要：目的辣椒素受体瞬时受体电位香草酸亚型1(transient receptor potential vanilloid 1,TRPV1)参与了多种肿瘤的生理和病理过程,但TRPV1在子宫内膜癌组织中的表达鲜见报道。本研究旨在观察子宫内膜癌组织及细胞中TRPV1的表达情况,并探讨其意义。方法收集河北医科大学第二医院2009-11-02-2015-08-31子宫内膜癌组织蜡块73例。对照组选取正常增殖期宫内膜10例;子宫内膜单纯或复杂性增生10例;子宫内膜非典型增生10例。免疫组化SP法测定TRPV1在不同类型子宫内膜癌组织中的表达,蛋白质印迹法检测人子宫内膜癌细胞系ISHIKAWA和HEC-1A的中TRPV1表达情况。结果免疫组化结果显示,TRPV1在子宫内膜癌组织的阳性表达率为42.5%(31/73),较正常内膜表达率100%(10/10)明显降低,χ~2=31.583,P<0.001。其中Ⅰ型子宫内膜癌组织的阳性表达率为73.5%(25/34),高于Ⅱ型子宫内膜癌的表达率15.4%(6/39),Z=-5.669,P<0.001。蛋白质印迹法结果显示,ER高表达的ISHIKAWA癌细胞系表达TRPV1蛋白,而ER低表达的HEC-1A癌细胞系未检测到TRPV1蛋白表达。结论Ⅰ型子宫内膜癌的发生发展可能有TRPV1参与,Ⅱ型子宫内膜癌的发生可能与TRPV1的丢失有关。子宫内膜癌中TRPV1的表达水平可能与ER的表达相关联。OBJECTIVE The capsaicin receptor TRPV1 participate in physiological and pathological processes of many tumors,but the expression of TRPV1 in endometrial carcinoma has not been reported.The purpose of this study was to investigate the expression of TRPV1 in endometrial carcinoma tissues and cells,and to explore its significance.METHODS 73 endometrial carcinoma wax blocks were from the Second Hospital of Hebei Medical University between 2009-11-02 and 2015-08-31.10 cases of normal proliferation phase endometrium,10 cases of simple or complex endometrial hyperplasia,10 cases of endometrial atypical hyperplasia were secleceted as the control group.Immunohisto-chemical SP method was used to examine the expression of TRPV1 in different types of endometrial lesion tissues,and western blot technique was used to examine the expression of TRPV1 in hunman endometrial carcinoma cell lines of ISHIKAWA and HEC-1A.RESULTS Immunohistochemical results showed that the positive rate of TRPV1 in endometrial carcinoma tissue was 42.5%（31/73）,which was much lower than in normal endometrium（100%,10/10）（χ^2=31.583,P〈0.001）.The expression rate of TRPV1 in typeⅠwas（73.5%,25/34）,which was higher than in typeⅡ（15.4%,6/39;Z=-5.669,P〈0.001）.Western blot results showed that the expression of TRPV1 was detected in ISHIKAWA cell lines in which the expression of ER was high,but it was not detected in HEC-1Acell lines which the expression of ER was low.CONCLUSIONS TRPV1 may participate in the occurrence and development of typeⅠendometrial carcinoma.The occurrence of typeⅡendometrial carcinoma may be associated with the lost of TRPV1.The expression level of TRPV1 in endometrial carcinoma may be associated with the expression of estrogen receptors.
