摘 要:为了开发从天然橡胶乳清中规模化提取白坚木皮醇技术,课题组以膜分离技术替代传统的蒸发浓缩和柱层析工艺,对天然橡胶乳清进行除杂和浓缩,结合结晶法提纯,制备出高纯度白坚木皮醇成品。试验采用截留分子量为1000 Da的超滤膜处理乳清,最大化的去除了杂质,接着用纳滤膜对乳清进行浓缩,去除小分子无机盐,经过膜处理的乳清浓缩液极易结晶,大大提高了生产效率,实现了白坚木皮醇的规模化提取。提取样品经检测,各项指标与白坚木皮醇特征参数相符。To develop a method for large-scale extraction of 1-quebrachitol from natural rubber serum. Membrane separa- tion technology was applied to concentrate and purify the natural rubber serum in the experiments, and the L-quebrachi- tol was extracted from the serum concentrate by crystallization. The crude extract was recrystallized three times from dis- tilled water, and then high purity sample was prepared by solventing-out process. The ultrafiltration (UF) membrane with a molecular weight cut-off (MWCO) of 1000 Daltons can reject most of the organic matters in the skim serum. The UF permeate was concentrated by nanofihration (NF) membrane, and L-quebrachitol was crystallized easily from the con- centrate. As a result, a new and convenient approach for large scale extraction of L-quebrachitol was proposed combining membrane separation and crystallization. In addition, 80g sample can be extracted for each operation with this approach under laboratory conditions. The structure of L-quebrachitol was confirmed by elementary analysis, infrared (IR) spec- troscopy,nuclear magnetic resonance (NMR) spectroscopy and mass spectroscopy (MS).
