不同阿胶酶解液相对分子量分布及补血升白作用对比研究

摘　　要：目的:制备阿胶及其酶解液,对比阿胶酶解前后相对分子量分布及补血、升白的作用。方法:采用TSKge L G2000SWXL(7.8 mm×300 mm,平均孔径512?),流动相为0.1 mo L/L Na CL+0.05%Na N3+0.1mo L/L磷酸缓冲液(p H=6.7),流速0.5 mo L/min,检测波长280 nm,对阿胶原液、阿胶猪胰脏粗酶酶解液、阿胶猪胰脏匀浆酶解液,匀浆空白液进行相对分子量分布测定;采用放血法致小鼠血虚模型及药物环磷酰胺所致小鼠白细胞减少模型,考察阿胶及其酶解液补血升白作用。结果:阿胶原液的蛋白质及多肽的相对分子质量主要区间为168 Da^1.7×105 Da,阿胶酶解液的相对分子量在3×103 Da^3×104 Da的区间内显著增多;阿胶酶解液可使失血所致血虚小鼠模型Hb、RBC水平显著提高,环磷酰胺所致小鼠白细胞减少模型的WBC、RBC、Hb水平显著提高。结论:阿胶酶解后相对分子质量减小,且酶解后补血升白效果更优;猪胰脏粗酶及匀浆物与胰蛋白酶的酶解效力相当。Objective：Preparation of donkey hide gelatin and its enzymatic hydrolysate, compared with donkey- hide gelatin before and after enzymolysis relative molecular weight distribution and the effect of enriching blood and promoting white.Method：GPC was performed on a TSK-geL G2000SWXL column （7.8 mm × 300 mm, average poresize 500A） with 0.05 moL/L phosphate buffered solution （pH6.7） at the flowrate of 0.5 mL/min, and detection at 280 nm.The Donkey hide gelatin, enzymatic hydrolysis of crude enzyme of pig pancreas with donkey hide gelatin, porcine pancreatic homogenate hydrolysate of donkey hide gelatin, homogenate blank solution relative determination of molecular weight distribution.the model of blood deficiency in mice induced by bloodletting and the model of mice induced by cyclophosphamide were studied.Result：The relative molecular weight of donkey hide gelatin solution is mainly in the range of 168 Da - 1.7 × 10^5 Da, the relative molecular weight of the enzymatic hydrolysate of donkey hide gelatin is mainly ranged from 3×10^3 Da to 3 × 10^4 Da.The enzyme hydrolysate of donkey hide gelatin can significantly improve the levels of Hb and RBC in mice with blood loss induced by blood loss, the levels of WBC, RBC and Hb in mice induced by cyclophosphamide were significantly increased.Conclusion：The relative molecular weight of the donkey hide gelatin decreased after enzymolysis, and the effect of blood enriching and white lifting is better after enzymolysis.The crude enzyme and homogenate of porcine pancreas have the same effect as trypsin hydrolysis.
