摘 要:目的研究不同剂量氯化汞对SD大鼠的血管内皮损伤情况。方法将54只雄性SD大鼠简单随机分为四组,高、中、低氯化汞中毒组,每组15只,对照组9只;采用高(17 mg·kg^(-1)·d^(-1))、中(8.5 mg·kg^(-1)·d^(-1))、低剂量(4.25 mg·kg^(-1)·d^(-1))浓度氯化汞盐水溶液灌胃建立SD大鼠亚急性汞中毒动物模型。中毒后7 d、14 d、21 d分别测定大鼠血细胞汞值、血清一氧化氮(NO)和内皮素-1(ET-1)值,同时行循环内皮细胞(CEC)计数。结果对照组血细胞汞值接近于零,中毒组与对照组差异均有统计学意义(P<0.05)。血清ET-1组间比较,高剂量组与对照组差异有统计学意义(P<0.05);组内比较,不同时期ET-1值差异有统计学意义(P<0.05)。血清NO组间比较,各剂量组与对照组差异均有统计学意义(P<0.05);同期比较,不同中毒剂量组之间比较差异也有统计学意义(P<0.05);组内比较,不同时期之间差异均有统计学意义(P<0.05)。血循环内皮细胞计数组间比较,各剂量组与对照组差异均有统计学意义(P<0.05);组内比较,不同时期之间差异均有统计学意义(P<0.05)。结论氯化汞可对血管内皮细胞有直接损伤作用,且损伤作用与剂量、时间存在同一变化趋势。Objective To study the injury of SD rats' vascular endothelial cell poisoned with different doses of mercuric chloride (HgC12). Methods The SD rat animal models were created by gavaging different doses of HgCI2. A total of 54 male rats were randomized into 4 groups: high, medium, low dose HgCI2 group (n= 15 in each, poisoned with 17 mg·kg^-1·d^-1, 8.5 mg-kg·^-1·d^-1, 4.25 mg·kg^-1·d^-1 HgC12, respectively), and control group (n=9). The levels of Hg in blood cell, NO and endothelin-1 (ET-1) in serum were measured on the 7^th day, 14^th day and 21^th day, and the number of circulating endothelial cells (CEC) was counted at the same time. Results Level of Hg: The levels of Hg in control group were close to zero, and there were statistically significant differences between high, medium, low dose HgCh groups and control group. Levels of serum ET-I: Between-group comparison showed significant differences between high dose HgCl2 group and control group, and within-group comparison revealed significant differences during different time. Levels of serum NO: Between-group comparison showed that there were significant differences between high, medium, low dose HgCh groups and control group, and also among the high, medium, low dose HgCI: groups at the same time, and with- in-group comparison revealed showed that there are significant differences during different time. CEC counts: Between-group comparison showed significant differences between high, medium, low dose HgCl2 groups and control group, and with- in-group comparison revealed significant differences during different time. Conclusion HgCl2 can cause direct injury and dysfunction to vascular endothelial cells, and its injury has the same changing trend with dose and time.
