摘  要:目的研究MAP3K2对A549细胞的增值、迁移和侵袭能力的作用。方法采用Micro RNA-520a-3p类似物不同程度下调非小细胞肺癌A549细胞MAP3K2水平,使用Western blot、MTT细胞增殖实验、划痕实验、Transwell实验等方法,对各组细胞的增殖、侵袭和迁移能力进行比较。结果通过细胞培养和mi RNA类似物干预,获得不同MAP3K2表达水平的A549细胞,并使用Western blot法进行验证,MTT细胞增殖实验、划痕实验和Transwell实验提示MAP3K2对A549细胞的增殖、侵袭和迁移能力具有双向调节作用。结论 MAP3K2对A549细胞的增值、迁移和侵袭能力具有双向作用,即先抑制后促进的过程。Objective To explore the effects of MAP3K2 on the proliferation, migration and invasion of A549 cells. Methods The analogue of MicroRNA-520a-3p was applied to the down regulation of the levels of MAP3K2 in non-small cell lung cancer (NSCLC) A549 cells to different degrees. The proliferation, invasion and migration ability of the cells in different groups were compared by such methods as Western blot, MTT cell proliferation test, scratch test and Transwell test. Results A549 ceils with different levels of MAP3K2 were obtained from cell culture and intervention of the analogue of miRNA. Western blot method was used for verification. MTT cell proliferation test, scratch test and Transwell test showed that MAP3K2 had bidirectional regulation effects on the proliferation, migration and invasion of A549 ceils. Conclusion MAP3K2 has bidirectional regulation effects on the proliferation, migration and invasion of A549 cells. It is a process of inhibition first and then promotion.
