摘 要:目的研究原癌基因人表皮生长因子受体2(HER2)对MCF10A人乳腺上皮细胞形态和迁移能力的影响及机制。方法通过慢病毒包装及感染MCF10A人乳腺上皮细胞,建立HER2过表达细胞,用TranswellTM法研究细胞迁移能力变化,结合Western blot法、实时定量PCR和免疫荧光染色检测细胞中上皮钙黏素(E-cadherin)的表达。结果过表达HER2的乳腺上皮细胞呈分散型生长,细胞迁移能力增强,E-cadherin蛋白表达受到抑制,细胞膜上E-cadherin表达也显著降低,重新过表达E-cadherin能抑制HER2诱导的细胞迁移和分散。结论 HER2能通过抑制E-cadherin促进人乳腺上皮细胞迁移。Objective To study the effect of HER2 on morphology and migration of MCF10 A human mammary epithelial cells and its mechanism. Methods The stable HER2-overexpressing MCF10 A cells were established by lentivirus infection.Then,cell migration was analyzed by TranswellTMassay; E-cadherin expression was detected by Western blotting,real-time quantitative PCR and immunofluorescent staining. Results HER2-overexpressing MCF10 A cells showed scattering growth pattern and increased migration ability. Overexpression of HER2 decreased the expression of E-cadherin protein in MCF10 A cells,and also reduced the membrance-associated E-cadherin. In addition,HER2-induced cell scattering and migration could be significantly suppressed by re-expression of E-cadherin. Conclusion HER2 promotes the migration of human mammary epithelial cells by inhibiting the expression of E-cadherin.
