摘 要:为了解河南省猪瘟病毒近2年的分子特征及变异情况,本试验使RT-PCR方法从猪瘟病料中获得了5株猪瘟病毒的E2基因全序列和其中10株E0基因全序列,并对猪瘟病毒阳性样品的E2和E0基因进行测序及变异分析。结果显示:扩增的河南省5株猪瘟病毒的E2基因,其中4株猪瘟病毒E2基因与石门株E2基因相比存在14个氨基酸的变化,1株和石门株E2基因存在4个氨基酸变化。与Shimen株相比,河南省猪瘟病毒的E2基因有1个糖基化位点的突变。扩增的10株猪瘟病毒的E0基因,发现河南省的猪瘟病毒之间E0基因同源性高,且其E0基因从密码子程度上一直在向远离经典毒株Shimen株和疫苗株HCLV的方向发展。对E2基因和E0基因进行进化分析发现河南省猪瘟病毒承受净化选择压力。以E2基因为基础进行分型,发现河南省共同存在猪瘟病毒1.1型和2.1b型;提示河南省猪瘟病毒变异种类更加多样化。In order to study the genetic variation of classical swine fever, We analyzed the E2 gene and E0 gene of CSFV strains isolated from domestic pigs in different farm of Henan province by RT-PCR during 2014-2015. Alignment of the E2 region of the 4 CSFV isolates with the Shimen strain showed 14 amino acid sequences substitutions,which were all at the same positions in the E2 gene. Another one had changes of 4 amino acid sequences. In addition,E2 gene of CSFV isolated Henan was analysed and discovered a mutation of glycosylation sites. Homology of E0 gene of CS- FV isolated Henan was extremely high,and far from the classical strains such as Shimen strain and vaccine strains HCLV. Furthermore, the positive selection pressures acting on the Erns and E2 en- velope protein genes of CSFV were assessed and a site-by-site analysis of the dN/dS ratio was per- formed to identify specific codons that undergo diversification under positive selection. While no significant evidence for positive selection was observed in E0 and E2. Phylogenetie analyses re vealed that all of the 5 CSFV isolates were clustered into genetic subgroup 1.1 and 2. 1b.
