摘  要:目的对黑果枸杞花青素的提取工艺进行响应面优化并检测其抗氧化活性。方法以黑果枸杞花青素的提取量为考察指标,采用4因素3水平的Box-Behnken实验设计优化黑果枸杞花青素的提取工艺参数并检测其抗氧化活性。结果在温度为50℃、料液比为1︰20、乙醇体积分数为50%的条件下提取3.0h时,花青素含量最高。黑果枸杞花青素的还原能力为64.3%;羟自由基的清除能力为60.5%;超氧阴离子的清除能力为87.2%;DPPH·的清除能力为78.3%。结论响应面法能够全面、准确地确定黑果枸杞花青素的提取工艺参数。黑果枸杞花青素在一定质量浓度范围内能够清除体内多余的自由基,具有一定的抗氧化活性,为黑果枸杞的功能研究奠定了基础。Objective To optimize the extraction process of anthocyanins from Lycium ruthenicum Murr.(black fruit wolfberry fruit),and to evaluate its antioxidant activity.Methods Response surface optimization was carried out.Taking the amount of anthocyanin extracted from black fruit wolfberry fruit as the research target,the Box-Behnken test was designed to optimize the extraction process parameters and the antioxidant activity of the extracts fromLycium ruthenicum Murr.with 4factors and 3levels.Results Under the conditions of temperature of 50 ℃,solid to liquid ratio of 1∶20,and the volume fraction of ethanol of 50%,extracting time of 3.0h,anthocyanin content was the highest with the reduction rate of 64.3%,the hydroxyl radical scavenging ability of 60.5%,the superoxide anion removal of 87.2%,and the ability of DPPH· clearance of 78.3%.Conclusion The response surface method can comprehensively and accurately determine the extraction process parameters of anthocyanin fromLycium ruthenicum Murr.anthocyanins fromLycium ruthenicum Murr.in a certain concentration range showed antioxidant activity,and can remove excess free radicals,which laid a foundation for further study.
