摘  要:目的分析内毒素血症肝损伤时肝组织局部血管紧张素1-7[Angiotensin 1-7,Ang(1-7)]及其特异性受体Mas受体表达变化。方法应用腹腔注射脂多糖(LPS)方法(10 mg/kg)制备内毒素血症急性肝损伤模型。雄性SD大鼠36只,随机分为对照组(C组,n=6只)、LPS组(2 h,6 h,12 h,24 h,48 h,共5组,每组6只)。LPS组经腹腔注射LPS,分别于2 h、6 h、12 h、24 h、48 h,用2%戊巴比妥麻醉大鼠,经心脏取血,取肝左叶组织;对照组注射生理盐水。qRT-PCR方法检测肝组织Mas受体mRNA的表达,Western-blot方法测定肝组织Mas受体蛋白表达水平,免疫组化方法测定Mas受体在肝组织分布情况,ELISA检测血清和肝组织匀浆中Ang(1-7)的含量。比较组间肝功能指标[谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)]、肝组织中Ang(1-7)及其受体Mas受体表达水平。采用SPSS 21.0统计软件进行单因素方差分析。结果 LPS注射后ALT、AST进行性升高,均于12 h达到峰值,而ALP于48h开始明显升高(P<0.01),48 h时肝组织病理损伤最重。与对照组相比,LPS注射后6 h肝组织中Mas受体蛋白开始增加,12 h肝组织中Mas受体mRNA开始升高,二者均于48 h达峰值(P<0.05);注射LPS后12 h肝组织匀浆中Ang(1-7)水平于48 h时显著升高(P<0.01)。结论 LPS注射48 h时肝组织病理损伤最重,此时肝脏局部Ang(1-7)及Mas受体表达最多,提示肝脏局部Ang(1-7)-Mas受体与内毒素血症急性肝损伤有一定相关性。Objective To study the expression of angiotensin 1-7 and its specific receptor Mas receptor in acute liver injury induced by lipopolysaccharide (LPS).Methods 36 male SD rats were randomly divided into control group (group C,n=6),LPS groups (2 h,6 h,12 h,24 h and 48 h groups,6 rats in each group).Rats in LPS groups were intraperitoneally injected with LPS (10 mg/kg).Anesthesia was achieved by intraperitoneal administration of 2% pentobarbital (1 mg/10 g body weight),then blood samples were drawn from heart,blood samples and tissue samples of left lobe of liver were collected in 2 h,6 h,12 h,24 h and 48 h respectively.The expression of mRNA of Mas receptor gene was detected by qRT-PCR and the expression of protein of Mas receptor in liver tissue was determined by Western blot.The analyses of data were performed by using SPSS version 21.0 and data were analyzed by ANOVA with comparisons among these groups.Results ALT and AST were significantly increased after injection of LPS,and reached peak at 12 h,while ALP began to increase significantly at 48 h (P&lt;0.01).The pathological damage of liver tissue was the most severe at 48 h.In comparison with that of control group,the protein expression of Mas receptor began to increase at 6 h after LPS injection,and the expression of mRNA of Mas receptor began to increase at 12 h,and both of them reached the peak at 48 h (P&lt;0.05) The levels of Ang (1-7) in liver tissue were significantly increased at 48 h (P&lt;0.01).Conclusion The expression of Ang (1-7) and Mas receptor in liver tissue at 48 h was the highest and the local Ang (1-7) and Mas receptor were associated with acute liver injury induced by LPS.
