摘 要:目的探讨CD29和CD49f对乳腺癌中肿瘤干细胞转移的影响。方法用流式分选技术分离CD24/CD29/CD49f亚群细胞。以伤口愈合、肿瘤细胞侵袭实验检测各标志物对肿瘤干细胞体外迁移分化的影响。以实时定量反转录聚合酶链反应(RT-qPCR)和免疫荧检测在裸鼠瘤分离得到的肿瘤细胞上的标志物。结果CD29和CD49f在癌症转移中具有重要作用,且两个蛋白的功能具有互补的特点。CD24^+/CD29^+双阳性细胞表达大量的间质型肿瘤细胞标志。CD24^-/CD29^-双阴性细胞则表达大量的上皮型肿瘤细胞标志。结论CD29和CD49f可能依赖于基质信号通路调节肿瘤干细胞间的基质连接并因此增强了肿瘤干细胞存活、分化和转移的能力。Objective In this study, we investigated the effect of CD29 and CD49f on metastatic potential of cancer stem cells (CSCs) isolated from mammary breast tumors.Methods We used flow cytometry to separated CD24^+ /CD29^+ /CD49f^+ characteristic cells. Meanwhile, we applied wound healing and tumor cell invasion experiment to detect the markers of CSC migration and differentiation. We used real-time reverse transcriptase-polymerase chain reaction(RT-qPCR) and immune fluorescence to detect markers of tumor cells separated from nude mice tumor. Results CD2^4+ CD29^+ cells maintained the ability to differentiate and reconstitute heterogeneity in the metastatic tumors whereas CD24^-CD29^- cells could not. Corresponding to their enhanced metastatic ability, CD24^+ CD29^+ cells exhibited features of the epithelial to mesenchymal transition.Conclusion Our findings indicate that CD29 and CD49f may regulate matrix connection between cancer stem cells through the substrate signaling pathways and enhance the survival, differentiation and transfer of cancer stem cell.
