摘 要:目的探究DACH2在肌层浸润性膀胱尿路上皮癌(UCB)中的表达及对预后判断的临床意义。方法通过免疫组化、Western Blot和RT-PCR技术检测伴淋巴结转移和不伴淋巴结转移的肌层浸润性UCB患者术后标本中的DACH2的表达情况,分析DACH2的表达及与患者预后之间的关系,验证DACH2能否作为独立预测肌层浸润性UCB预后的分子标记物。结果免疫组化、Western Blot和RT-PCR结果一致显示,DACH2在伴淋巴结转移的肌层浸润性UCB标本中有较高表达,且差异均有统计学意义(P<0.05)。DACH2表达与肿瘤高分级相关,差异有统计学意义(P<0.05),DACH2表达与患者无转移生存率和总生存率相关,差异有统计学意义(P<0.05)。结论 DACH2可作为一个有潜力的分子标记物指导肌层浸润性UCB预后的判断。Objective To investigate the expression of DACH2 in the muscle-invasive urothelial carcinoma of bladder(UCB) with lymph node metastasis and its clinical significance in predicting the prognosis of UCB. Methods The expression of DACH2 in the postoperative specimens of bladder cancer patients with lymph node metastasis and without lymph node metastasis was detected by immunohistochemistry, Western Blot and RT-PCR. The relation between the expression of DACH2 and the prognosis of bladder cancer patients was analyzed. And further the statistical analysis of clinical followup data was used to verify whether DACH2 can be an independent biomarker in predicting the metastasis and prognosis of muscle-invasive UCB. Results Immunohistochemistry, Western Blot and RT-PCR showed that DACH2 was highly expressed in the muscle-invasive bladder urothelial carcinoma with lymph node metastasis(P〈0.05). Analysis of the clinicopathology features of UBC showed that the expression of DACH2 was correlated with the high grade of UBC(P〈0.05). Analysis of clinical follow-up survival data revealed that DACH2 expression was associated with metastasis-free survival and overall survival. Conclusions DACH2 expression in bladder cancer patients with lymph node metastasis was higher than that in patients without lymph node metastasis. DACH2 can serve as a helpful and prognostic biomarker in muscle-invasive UCB.
