益气解毒法对胃癌细胞增殖活性影响

摘　　要：目的:观察益气解毒法对胃癌细胞增殖活性的干预作用。方法:空白血清、复方环磷酰胺含药血清、不同剂量(低剂量、中剂量和高剂量)益气解毒法含药血清,分别与人胃癌细胞SGC-7901共培养24、48、72、96 h,MTT法测其对胃癌细胞增殖的抑制率;流式细胞技术测含药血清与人胃癌细胞SGC-7901共培养48 h后的细胞周期。结果:益气解毒法含药血清依照剂量从低到高的顺序,其吸光度(OD)值依次降低并与刺激时长负相关,抑制率(IR)依次升高并与刺激时长正相关;在24~72 h抑制强度随时间逐渐增强,72~96 h抑制强度略有降低。益气解毒法低、中、高剂量各组含药血清在分别刺激胃癌SGC-7901细胞48 h后,其G0/G1期细胞比率显著增加(P<0.05),并与益气解毒法的剂量正相关;S期和G2/M期细胞比率均显著减少(P<0.05),并与益气解毒法的剂量负相关。结论:益气解毒法对胃癌细胞增殖有抑制作用。Objective：To observe intervention effect of nourishing Qi and relieving poison method on gastric cancer cell proliferative activity. Methods：Human gastric cancer cells SGC-7901 were cultured respectively with the rat serums containing nourishing Qi and relieving poison method at different doses（low-dose,mid-dose and high dose）to the indicated time points（24 h,48 h,72 h and 96 h）,blank rat serums as well as rat serums containing compound cyclophosphamide as negative and positive control respectively,whose inhibition rates on SGC-7901 cell proliferation were detected by MTT. When human gastric cancer SGC-7901 cells were cultured respectively with above-mentioned medicated serums for 48 h,their cell cycles were detected by flow cytometry. Results：The optical densities（OD）of human gastric cancer SGC-7901 cells cultured with the rat serums containing nourishing Qi and relieving poison method decreased sequentially according to from the low-dose to high dose,and there was a negative correlation between their OD and their stimulation length,but their inhibition rates（IR）on SGC-7901 cells increased sequentially and positively correlated with their stimulation length;During 24-72 h the inhibition intensities of the rat serums containing nourishing Qi and Relieving Poison method on SGC-7901 cells gradually increased with the increasing stimulation time,but from 72-96 h the inhibition intensities showed slight decrease. After SGC-7901 cells were cultured for 48 h with the rat serums containing nourishing Qi and relieving poison method at different doses,the G0/G1 stage ratios showed a significant increase（P〈0.05）,a positive correlation with the doses of nourishing Qi and relieving poison method;The S stage and G2/M stage ratios showed a significant decrease（P〈0.05）,a negative correlation with the doses of nourishing Qi and relieving poison method. Conclusion：Nourishing Qi and relieving poison method can inhibit gastric cancer cell proliferative activity.
