逍遥散对肝郁气滞证视神经炎大鼠视路NR1表达研究

摘　　要：目的:研究逍遥散对肝郁气滞证视神经炎大鼠视路外侧膝状体、视皮质NR1表达的影响。方法:将60只大鼠按RAND函数随机分为正常组、肝郁气滞证视神经炎组(病证结合组)、逍遥散加肝郁气滞证视神经炎组(给药组)3组。采用EAE动物模型方法建立视神经炎大鼠模型,同时应用慢性束缚应激方法增加肝郁气滞因素,最终形成肝郁气滞证视神经炎大鼠模型。观察大鼠行为学,记录旷场实验,利用上述指标评价模型造模成功后,免疫组化法检测各组大鼠外侧膝状体、视皮质NR1的动态变化。结果:逍遥散能降低EAE神经功能评分,旷场实验总分降低,病证结合组外侧膝状体、视皮质NR1表达增高,给药组表达降低。结论:逍遥散可以降低肝郁气滞证视神经炎大鼠视路NR1的表达,可调节肝郁气滞证视神经炎大鼠的功能紊乱。Objective：To study the impact of Xiaoyao San on liver depression and Qi stagnation type optic neuritis in the visual pathway of rat lateral geniculate body,NR1 expression of visual cortex. Methods：60 rats were randomly divided into control function RAND group,liver depression and Qi stagnation type optic neuritis group,Xiaoyao San group liver depression and Qi stagnation type optic neuritis three groups,chronic restraint stress application method of established liver qi stagnation animal model,the type with EAE animal model method to establish animal model of optic neuritis,the rats behavior,open-field test,after the use of the index evaluation model successful model,immunohistochemical method to detect the lateral geniculate body,depending on the dynamic changes of cortical NR1. Results：Xiaoyao San could reduce EAE neurological score significantly lower open field test scores,liver depression and Qi stagnation type group could depend on the lateral geniculate body neuritis,visual cortex NR1 expression increased,decreased expression of the administration group. Conclusion：Xiaoyaosan San reduce the expression of liver depression and Qi stagnation type optic neuritis in the visual pathway of rats NR1,adjustable liver depression and liver depression and Qi stagnation type type rats with optic neuritis dysfunction.
