摘 要:目的:观察活性维生素D3腹腔注射对2型糖尿病大鼠海马神经元的影响。方法:SD大鼠30只,随机分为空白对照组(Control),高脂组(HF),高脂+VD组(HF+VD),糖尿病组(DM),糖尿病+VD组(DM+VD)。测定大鼠空腹血糖、胰岛素、维生素D等生化指标,应用透射电镜观察海马神经元结构变化。结果:与对照组相比,HF组、HF+VD组大鼠体重增加,(分别为343.28±10.41 g;356.81±36.20 g和360.18±25.56g),DM组体重降低(265.6±16.11 g)(P<0.05);DM组SDF-1为398.33±15.01 ng/L,低于Control组(457.53±26.56 ng/L),骨化三醇干预后SDF-1显著升高(448.54±36.83 ng/L)(P<0.05);DM组(25-OH-D)水平为165.25±30.75μg/L,低于Control组213.65±30.79μg/L,补充骨化三醇后显著提高至210.31±50.69μg/L(P<0.05);DM组海马神经元CXCR4表达降低,线粒体可见明显肿胀,DM+VD组线粒体形态明显改善,海马神经元CXCR4表达水平明显升高。结论:常规量补充维生素D可提高糖尿病大鼠体内维生素D水平,对海马神经元有明显保护作用。Objective:To explore the effect of vitamin D3 on the hippocarpal neurons of type 2 diabetic rats.Methods:30 female SD rats were randomly divided into control group,high fat group (HF),high fat + vitamin D (HF + VD),diabetes mellitus group (DM),diabetes mellitus + vitamin D (DM + VD).The blood sugar,insulin and vitamin D level were detected at the beginning and end of the experimental period.The hippocampal neurons were observed by transmission electron microscope.Results:Compared with control group (343.28± 10.41 g),body weight of HF (356.81± 36.20 g),HF + VD rats (360.18± 25.56 g) increased and that of DM rats (265.6± 16.11 g)decreased(P〉0.05).SDF-1 decreased in diabetic rats(398.33± 15.01 ng/L).calcitriol treatment reversed the changes significantly (448.54± 36.83 ng/L)(P〈0.05).25-OH-D decreased in DM rats (165.25± 30.75 μg/L)and calcitriol treatment increased 25-OH-D in DM rats 210.31 ± 50.69 μg/L (P〈0.05).The expression of CXCR4 decreased in hippocampal neurons of DM rats,and swelled mitochondria was observed in the neurons.calcitriol treatment improved the injury.Conclusion:calcitriol treatment increased vitamin D level,and has protective effect on hippocampal neurons in type 2 diabetes mellitus rats.
