唑来膦酸联合IL-2体外诱导胃癌患者外周血γδT细胞的研究

摘　　要：目的:探讨唑来膦酸联合白细胞介素-2(IL-2)诱导胃癌患者外周血γδT细胞的作用。方法:收集胃癌患者和健康捐献者各30例,分别采集外周静脉血并分离出单核细胞(PBMC),采用流式细胞仪(FCM)检测唑来膦酸联合IL-2诱导前后γδT细胞的数量及其CD69分子的表达情况。结果:胃癌组的γδT细胞数量和百分比均明显低于健康对照组(P<0.05);唑来膦酸联合IL-2诱导PBMC后,胃癌实验组和健康对照组中的γδT细胞的数量和百分比均较诱导前明显升高(P<0.05)。结论:唑来膦酸联合IL-2体外诱导PBMC可显著扩增γδT细胞,以加强杀伤肿瘤细胞的目的。Objective：To determine the effects of zoledronate plus interlukin 2 （IL-2） on the proliferation of peripheral blood mononuclear cells （PBMCs） of patients with gastric cancer in vitro.Methods：PBMCs were isolated from 30 healthy doners and 30 patients with gastric cancer respectively,and then stimulated with zoledronate plus IL-2 in vitro.Flow cytometry （FCM） was used to evaluate the γδT cell subpopulation of PBMCs and detect the expression of CD69 in γδT cells.Results：The percentage and absolute number of γδT cells from PBMCs were both lower in patients with gastric cancer than those of the control group （P〈0.05）.The combination of zoledronate and interlukin 2 could significantly increase the percentage and absolute number ofγδT cells from PBMCs in patients with gastric cancer and healthy control group （P〈0.05）.Conclusion：Zoledronate and IL-2 could obviously stimulate the proliferation of γδT cells from PBMCs to further kill the cancer cells.
