摘 要:本研究旨在探析IGF2、H-FABP和RPL9基因在猪不同组织和两种猪间的表达差异。选用体重70 kg左右的杜×长×大三元杂交猪和豫南黑猪,分别采集其心、肝、脾、肺、肾、腹部脂肪和背部皮下脂肪,进行总RNA提取及不同基因的荧光定量PCR实验。结果表明:在豫南黑猪心脏、肝脏和脾脏组织中IGF2基因的转录水平约为三元杂交猪的225.0倍(P<0.05);豫南黑猪心脏、脾脏和皮下组织中H-FABP基因的转录水平高于三元杂交猪,其中脾脏组织H-FABP的表达量约为三元杂交猪的6.8倍(P<0.05),而三元杂交猪肺脏和腹部脂肪H-FABP表达量约为豫南黑猪的2.7倍(P<0.05);RPL9基因在豫南黑猪心、肝、肺和肾组织中表达量高于三元杂交猪,在脾脏、皮下脂肪和腹部脂肪中表达量低于三元杂交猪内表达量,其中皮脂中的表达量差异最大,三元杂交猪约为豫南黑猪的4.9倍(P<0.05)。综上可知,两种猪不同组织间IGF2、H-FABP和RPL9基因的表达差异显著。This experiment was conducted to study the differentially expressions of IGF2, H-FABP and RPL9 genes in different tissues between the yunan black pig and the three breed hybrid pig and analyzed the differences of gene expression between different pigs. The yunan black pig and the" Duroc large white x Landrace" three breed hybrid pig, weighing an average of 70 kg, were randomly chosen to obtain the heart, liver, spleen, lung, kidney, abdominal fat and back subcutaneous fat for total RNA extraction and fluorescence quantitative PCR test of the IGF2, H-FABP and RPL9 gene. The results showed that the expression differences of IGF2 in heart, liver, spleen tissues of yunan black pig were especially obvious that it of the yunan black pig were about 225.0 times of the three breed hybrid pig (P〈0.05). The transcription levels of H-FABP in the yunan black pig heart, spleen, and subcutaneous tissues were higher than those in the three breed hybrid pig, which the expression in spleen tissue of the yunan black pig was about 6.8 times of the three breed hybrid pig (P〈0.05), while the expressions in lung and abdomen fat of the three breed hybrid pig were higher, that H-FABP expression in abdomen fat of the three breed hybrid pig was about 2.7 times of the yunan black pig (P〈0.05). The expressions of RPL9 in heart, liver, lung and kidney tissues of the yunan black pig were higher and the expression difference in subcutaneous fat was largest, which of .the three breed hybrid pig was about 4.9 times that of the yunan black pig (P〈0.05). These results indicated that the expression differences of the IGF2, H-FABP and RPL9 gene in different tissues between the yunan black pig and the three breed hybrid pig were significant.
