药用植物广藿香内生真菌的生物活性研究

摘  要:目的:研究40株广藿香内生真菌的生物活性,为进一步发掘活性代谢产物提供理论依据。方法:采用SRB、PNPG和DPPH法分别测定内生真菌提取物的细胞毒、抑制α-葡萄糖苷酶和清除DPPH自由基等活性。结果:21株内生真菌对肿瘤细胞SF-268、MCF-7、NCI-H460、Hep G-2中的至少3种细胞具有抑制活性,IC_(50)值在0.03~20.30μg/mL之间,其中菌株A616的抑制活性最为显著,对4种肿瘤细胞的IC_(50)值分别为0.044、0.031、0.030、0.032μg/mL;13株内生真菌具有抑制α-葡萄糖苷酶活性,其中菌株A598的活性最为显著,其IC_(50)值为184.38μg/mL;15株内生真菌具有DPPH自由基清除活性,其中菌株A594、A598、A630显示出较强的DPPH自由基清除能力,IC_(50)值分别为36.33、20.63、59.93μg/mL。结论:广藿香内生真菌具有多种生物活性,值得进一步深入研究。Objective To investigate biological activities of 40 endophytic fungal strains derived from Pogostemon cabin, thus providing a theoretical basis for further searchfor biologically activemetabolites. Methods Cytotoxic, α - glueosidaseinhibitoryand DPPH free radical scavenging activities of endophytic fungal extractswere examined by the SRB, PNPG and DPPHmethods, respectively. Results 21 strains showed inhibitory activity against at least three tumor cell lines of SF - 268, MCF - 7, NCI - H460 and HepG - 2 with IC50 values ranging from 0. 03 to 20. 30μ/mL. Among them, the strain A616 exhibited the strongest eytotoxicity against the four tumor cell lines with IC50 values of 0. 044, 0. 031,0. 030 and O. 032 lag/mL, respectively. 13 strains showed α- glucosidaseinhibitory activity, among which the strain A598 displayed the strongest inhibitory activity with an IC50 of 184. 38μg/mL. 15 strains showed DPPH free radical scavenging activity, and among them the strains A594, A598 and A630 exhibited the stronger free radical scavenging capacity with IC50 of 36. 33, 20. 63 and 59. 93 μg/mL, respectively. Conclusion Endophytic fungi derived from P. cabin showed several biological activities and deserved further study.
