摘  要:目的:探讨铁皮石斛多糖(dendrobium polysaccharides,DP)对高糖诱导的SD大鼠胸主动脉内皮依赖性舒张功能(endothelium dependent relaxation,EDR)的影响。方法:采用离体血管灌流方法,观察不同浓度DP对高糖诱导的大鼠胸主动脉EDR的影响。Western Blot法检测葡萄糖调节蛋白78(GRP78)和C/EBP同源蛋白(CHOP)及核转录因子-κB(NF-κB)的表达。结果:高糖(44 mmol·L^(-1))显著抑制了血管环EDR,而对非内皮依赖性舒张功能没有影响;DP(200,400和800μg·mL^(-1))逆转了高糖对EDR的抑制作用。高糖显著增加胸主动脉中GRP78和CHOP及NF-κB的表达(P<0.05)。与高糖组比较,高糖+DP(200,400和800μg·mL^(-1))组GRP78和CHOP及NF-κB的表达均显著降低(P<0.05)。结论:DP对高糖诱导的EDR抑制具有保护作用,其机制可能与抑制内质网应激和NF-κB信号通路有关。Objective: To investigate the effect of dendrobium polysaccharides( DP) on the injury of endothelium dependent relaxation( EDR) induced by high glucose of isolated thoracic aorta in Sprague-Dawley( SD) rats and explore the mechanism. Methods: Organ baths in thoracic aortic rings of SD rats were used to investigate the effect of different concentrations of DP on the injury of EDR of thoracic aorta rings induced by high glucose. The expressions of endoplasmic reticulum stress protein markers including glucose-regulated protein 78( GRP78),CCAAT/enhancer-binding protein homologous protein( CHOP) and nuclear transcription factor kappa B( NF-κB) in the thoracic aorta rings were measured by Western Blot. Results: High glucose( 44 mmol·L^-1)inhibited significantly EDR mediated by acetylcholine in thoracic aorta rings of rats. High glucose did not affect non-EDR mediated by sodium nitroprusside in thoracic aorta rings of rats. The injury of EDR in thoracic aorta rings of rats induced by high glucose was inhibited by DP( 200,400 and 800 μg·mL^-1). High glucose increased significantly the expressions of GRP78,CHOP and NF-κB in the thoracic aorta rings( P〈0. 05). Compared withthe high glucose group,the expressions of GRP78,CHOP and NF-κB in the thoracic aorta rings in high glucose +dendrobium polysaccharides( 200,400 and 800 μg·mL^-1) group were significantly decreased( P〈0. 05).Conclusion: Dendrobium polysaccharides inhibit the injury of EDR of thoracic aorta rings induced by high glucose,and the mechanisms may be related with the inhibition of endoplasmic reticulum stress and NF-κB signaling pathway.
