摘  要:目的:研究HO-1抑制剂锌原卟啉IX(ZnPPIX)对胃癌细胞增殖能力和侵袭能力的影响,探讨锌原卟啉对胃癌细胞生物学特性的调控作用。方法:应用胃癌细胞株SGC7901作为实验载体,不同浓度ZnPPIX干预胃癌细胞,应用MTT法检测其在不同时间段和不同浓度对胃癌细胞增殖能力的影响。应用Transwell小室检测ZnPPIX对胃癌细胞侵袭能力的影响。Western blot在蛋白水平检测胃癌细胞中侵袭相关因子的表达变化。结果:ZnPPIX可明显抑制肿瘤细胞的增殖能力且呈浓度依赖性。ZnPPIX干预胃癌细胞与Co PP组和空白对照组相比,细胞的侵袭能力明显减弱。ZnPPIX可降低胃癌细胞MMP2、MMP9、VEGF的蛋白表达量。结论:本实验初步探讨HO-1抑制剂锌原卟啉IX对胃癌细胞增殖和侵袭能力的负向调控作用,为HO-1可能成为胃癌的治疗新靶点提供理论依据及线索。Objective: To study the potential effect of protoporphyrin Ⅸ zinc( ZnPPIX) which was heme oxygenase-1( HO-1) inhibitor on the proliferation and invasion ability of gastric cancer cells. Investigate the regulation effect of ZnPPIX on biological characteristic in gastric cancer cells. Methods: Gastric cancer cells SGC7901 was used in vitro,which were intervened by ZnPPIX with different concentrations. MTT assay was carried to examine the effects of ZnPPIX on the proliferation ability in gastric cancer cells in different concentrations and at different time points.The effect of ZnPPIX on the invasion ability of gastric cancer cells were assessed by using Transwell chamber. The expression of invasive factors in gastric cancer cells were measured by Western blot. Results: The proliferation ability of gastric cancer cells was significantly enhanced by ZnPPIX by a concentration-dependent manner. ZnPPIX significantly reduced the invasion ability of gastric cancer cells,compared with Co PP or the control group. The protein expression of MMP2,MMP9,and VEGF in gastric cancer cells was significantly inhibited by ZnPPIX. Conclusion: This experiment suggests the negative regulation effect of ZnPPIX on the proliferation and invasion ability of gastric cancer cells. Our research provides the theoretical basis and clues for HO-1 being a new therapy target in gastric cancer.
