摘　　要：目的:探讨LncRNA MEG3在胶质瘤组织标本中的表达及与临床病理特征之间的关系,探讨其在预测胶质瘤患者预后中的价值。方法:采用QRT-PCR法检测92例胶质瘤组织标本和48例正常脑组织中的LncRNA MEG3表达,分析其表达与患者临床病理特征及预后的关系。结果:LncRNA MEG3在胶质瘤组织标本中的表达较正常脑组织明显降低;LncRNA MEG3的表达与WHO分级(P=0.002)、Karnofsky评分(KPS)(P<0.001)、复发时间(P=0.001)、肿瘤大小(P=0.032)和生存时间(P=0.009)显著相关;Kaplan-Meier分析结果表明,LncRNA MEG3低表达患者的总生存时间(OS)(P<0.001)和无进展生存期(PFS)(P<0.001)较高表达者明显缩短,差异具有统计学意义。多因素分析显示胶质瘤中LncRNA MEG3的低表达是预测OS(HR:4.053;95%CI:5.820~17.680;P=0.001)和DFS[HR:3.140;95%CI:2.468~10.820;P<0.001]的独立危险因素。结论:LncRNA MEG3的表达与胶质瘤患者预后显著相关,LncRNA MEG3可作为预测胶质瘤患者预后的独立标志物。Objective：To investigate the relationship between LncRNA MEG3 expression and clinicopathologic factors in glioma tissues and evaluate the role of LncRNA MEG3 expression in predicting the prognosis of glioma patients.Methods：The expression levels of LncRNA MEG3 were analyzed in 92 gliomas tissues and 48 normal brain tissues by quality real-time PCR（QRT-PCR）.Survival analysis by Kaplan-Meier method was performed to assess prognostic significance.Results：Down-regulation of LncRNA MEG3 was detected in most primary glioma tissues,which was confirmed by QRT-PCR analysis.Additionally,down-regulation of LncRNA MEG3 was significantly associated with WHO grade（P=0.002）,Karnofsky performance score（KPS）（P〈0.001）,time to recurrence（P=0.001）,tumor size（P=0.032） and survival time（P=0.009）.Kaplan-Meier analysis showed that high expression of LncRNA MEG3,the overall survival time（OS）（P〈0.001） and progression free survival（PFS）（P〈0.001） were significantly shorter than high expression of LncRNA MEG3.COX regession analyses showed that LncRNA MEG3 expression was the independent risk factors for predicting OS（HR：4.053;95% CI：5.820-17.680;P=0.001） and DFS[HR：3.140;95% CI：2.468-10.820;P〈0.001] of glioma patients.Conclusion：LncRNA MEG3 expression is significantly correlated with the prognosis of glioma patients,suggesting that LncRNA MEG3 may serve as the independent prognostic marker.
