电针“内关”预处理对缺血再灌注大鼠心肌线粒体呼吸功能的影响

摘　　要：目的：探讨电针＂内关＂预处理对心肌缺血再灌注损伤过程中心肌线粒体的保护作用。方法：本实验选用3月龄Wistar雄性大鼠45只,体重180-220g,编号后按随机数字表分入空白组、模型组、电针预处理组,每组15只。造模采用结扎左冠状动脉前降支,缺血30分钟,再灌注2小时,观察心肌线粒体的呼吸功能及电镜下超微组织的变化。结果：电针预处理组大鼠部分心肌肌丝溶解、断裂,心肌细胞轻微肿胀,轻度空化,线粒体排列稍紊乱,部分线粒体水肿、存在空泡样病变。电针预处理组的大鼠线粒体RCR处于正常水平,明显高于模型组,存在显著性差异（P〈0.01）,但电针预处理组线粒体RCR低于空白组,差异显著（P〈0.01）。结论：电针＂内关＂预处理能改善心肌组织形态学表达,防止心肌组织结构的破坏,修护心肌缺血性损伤,更对线粒体的结构与功能起到了良性的保护作用。Objective ： To explore the protective effects of electroacupuncture preconditioning at ＂Neiguan＂ （ PC6 ） on myocardial cells in the process of myocardial ischemia and trperfusion injury. Methods ： In this experiment,45 male Wistar rats, 3 - month - old, weighing about 180 -220 g, were randomly divided into three group：no -treatment control group （NC group） , myocardial ischemia -reperfnsion injury model group （M group） and electroacupuncture preconditioning group （EA group）, with 15 rats in each group respectively. Ischemia - reperfusion injury model was achieved by the ligation and deligation of left anterior descending artery with ischemia for 30 rain and reperfusion for 120 rain. Before establishing the model, the EA group with electroacupuncture for 20 rain, 1 mA,continuous wave. And then the myocardial tissue was taken out and changes of the respiratory control ratio of mitochondria,the content of ATP and the ultrastructures were observed. Results ： In the EA group, the structure of cardiac tissue was normal except for a few mild edemata among cells, interstitial and mitochondria. The RCR of mitochondria in the EA group was in the normal level and significant higher than that in the M group （ P 〈 0. 01 ）, but lower than that in the NC group （ P 〈 0. 01 ）. Conclusion：Electroacupuncture preconditioning at ＂Neiguan＂ （PC6） could repair the damage causing by ischemia - reperfusion injury in myocardial tissue and play a positive role in the protection of structure and function of mitochondria.
