小鼠皮肤心脏联合移植模型的建立及移植物功能监测

摘　　要：目的探讨小鼠皮肤心脏联合移植模型的建立及移植物功能的监测。方法以BALB/C小鼠、C57BL/6小鼠做为供、受者,单纯皮肤移植组采用尾-背法行皮肤移植,单纯心脏移植组采用腹腔异位心脏移植法行心脏移植,皮肤心脏联合移植组序贯采用尾-背皮肤移植及腹腔异位心脏移植模型。术后采用腹部触诊的方法,监测心脏移植物功能,通过观察残存移植皮片大小,监测皮肤移植物功能。结果皮肤心脏联合移植组皮肤移植物存活时间较单纯皮肤移植组略有延长(皮肤心脏联合移植物v.s.单纯皮肤移植组,8天v.s.7天),但两组皮肤移植物存活时间比较没有显著差别(P=0.1290)。单纯心脏移植组心脏移植物和皮肤心脏联合移植组心脏移植物平均存活时间均为9天(MST=9天),两组心脏移植物存活时间无显著差别(P=0.6077)。结论小鼠皮肤心脏联合移植模型稳定,移植物功能监测简单方便,适用于移植免疫学方面的研究。Objective To explore the technique of mouse skin plus heart transplantation and the function of graft monitoring.Methods Methods：BALB/C and C57BL/6mice were used as donors and recipients,respectively.Skin only group：C57BL/6recipients received tail skin from BALB/C donors.Heart only group：C57BL/6recipients received heart grafts from BALB/C donors.Skin plus heart group：C57BL/6recipients received heart and tail skin grafts from BALB/C donors at the same time.Heterotopic palpation was performed to monitor the function of heart graft after transplantation,the function of skin grafts were monitored by observing the size of surviving grafts.Results Median survival time（MST）of skin grafts in skin only group was 7days,the skin grafts survival time have slightly longer compared to skin only group,but there were no significant difference（MST=8days,P=0.1290）.There were no significant difference of heart grafts survival time in both heats only group and skin plus heart group（MST=9days,P=0.6077）.ConclusionSkin plus heart transplantation procedure is stable to perform.Heterotopic palpation of hearts grafts and observing the size of skin grafts are simple to monitor the function of grafts.The skin plus heart transplantation model by using this procedure and monitoring protocol is useful for immunological research related to transplantation.
