摘 要:目的探讨健脾消积汤含药血浆对肝细胞癌HepG2细胞的抑制作用及其机制。方法将30只SD大鼠随机分为健脾消积汤组及空白对照组,每组15只,分别以健脾消积汤、生理盐水灌胃后,获取其血浆。以10%、15%、20%健脾消积汤组大鼠血浆作用于肝癌HepG2细胞(含药血浆组),同时以等浓度的空白对照组大鼠血浆处理HepG2细胞(空白血浆组)。两组细胞分别培养24 h、48 h、72 h后,以CCK-8法检测HepG2细胞的增殖情况。采用两组大鼠15%血浆作用于肝癌HepG2 48 h后,检测细胞的迁移情况及其转化生长因子-β_1(TGF-β_1)的mRNA、蛋白表达水平。结果不同浓度空白血浆组及含药血浆组的A值比较,差异均有统计学意义(P<0.05),其中15%、20%含药血浆组在48 h的A值均低于相对应浓度的空白血浆组(P<0.05);含药血浆组仅于48 h时对HepG2细胞增殖的抑制率>0,但各组间的抑制率比较差异无统计学意义(P>0.05)。含药血浆组HepG2的迁移细胞数少于空白血浆组(P<0.05)。含药血浆组TGF-β_1mRNA的相对表达量低于空白血浆组(P<0.05),且TGF-β_1的蛋白表达量较空白对照组有所下调。结论健脾消积汤含药血浆对肝癌HepG2细胞增殖、迁移有一定的抑制作用,其机制可能与下调TGF-β_1的表达有关。Objective To explore the inhibitory effect of Jianpi Xiaoji Tang drug-containing plasma on hepatocellular carcinoma HepG2 cells and its mechanism. Methods Thirty SD rats were randomly divided into Jianpi Xiaoji Tang group and blank control group,with 15 rats in each group. Intragastric administration of Jianpi Xiaoji Tang and normal saline were separately performed in the corresponding group,then the plasma of rats was collected. Hepatoma HepG2 cells were treated with 10%,15% and 20% rat plasma of Jianpi Xiaoji Tang group(drug-containing plasma group),while other HepG2 cells were treated with the same concentrations of rat plasma of blank control group(blank plasma group). At 24,48 and 72 hours after culture,the proliferation of the HepG2 cells in both groups were determined by cell counting kit-8(CCK-8) test. Hepatoma HepG2 cells were treated with 15% rat plasma of both groups for 48 hours,then the migration of cells,mRNA and protein expression levels of transforming growth factor-β_1(TGF-β_1) were detected. Results There was significant difference in A value among the blank plasma groups and drug-containing plasma groups with different concentrations(P〈0. 05),and the A values of the15%,20% drug-containing plasma groups were lower than the values of the blank plasma groups with corresponding concentrations at 48 hours(P〈0.05). The inhibitory rates of drug-containing plasma groups on HepG2 cells proliferation were more than 0 only at 48 hours,but no significant difference in the inhibitory rate was observed(P〉0. 05). The number of migrating HepG2 cells in the drug-containing plasma groups was less than that in the blank plasma groups(P〈0. 05). In the drug-containing plasma groups,the relative expression of TGF-β_1 mRNA decreased compared to the blank plasma groups(P〈0. 05),and the expression of TGF-β_1protein was down-regulated compared to the blank control group.
