摘 要:为了探讨骨髓间充质干细胞(BMMSCs)对小鼠实验性变态反应性脑脊髓炎(EAE)的治疗作用,将36只小鼠随机分为3组,每组12只,分别为对照组、模型组和BMMSCs组.采用不同方法治疗8周后,通过反转录PCR(RTPCR)和蛋白质免疫印迹(Western blot)分别检测小鼠脊髓组织中白介素-1(IL-1)、IL-2、IL-4、IL-6、IL-10、IL-12、肿瘤坏死因子-α(TNF-α)和干扰素-γ(IFN-γ)的mRNA水平和蛋白水平.结果显示:与对照组比较,模型组IL-1、IL-2、IL-12、TNF-α和IFN-γ的mRNA水平和蛋白水平上调(p<0.05),而IL-4和IL-10下调(p<0.05);与模型组比较,BMMSCs组IL-1、IL-2、IL-12、TNF-α和IFN-γ的mRNA水平和蛋白水平下调(p<0.05),而IL-4和IL-10上调(p<0.05).进而用苏木精-伊红(HE)染色观察小鼠脊髓组织形态学改变,结果显示:对照组脊髓组织的结构正常,神经细胞形态完好;模型组脊髓组织内出现大量炎性细胞;BMMSCs组脊髓组织内没有发现明显的炎性细胞.由上述结果可知,BMMSCs对EAE小鼠具有治疗作用,其机制与抑制炎性因子的表达和减少变态反应性脑脊髓炎对脊髓的损伤有关.To study the treatment effects of bone mesenchymal stem cells(BMMSCs)on the mice allergic encephalomyelitis,36 mice were randomly divided into three groups(control group,model group and BMMSCs group),12 for each group,and treated with different methods.After treatment for 8 weeks,expressions of IL-1,IL-2,IL-4,IL-6,IL-10,IL-12,TNF-α and IFN-γ of spinal cord tissues in mice were analyzed by RT-PCR and Western blot,and HE staining were performed to observe morphological changes of spinal cord tissues in mice.The RT-PCR and Western blot results showed as follows:compared to the control group,mRNA and protein levels of IL-1,IL-2,IL-12,TNF-α and IFN-γ significantly increased(p〈0.05),while those of IL-4 and IL-10 significantly decreased(p〈0.05)in the model group.In the BMMSCs group,the mRNA and protein levels of IL-1,IL-2,IL-12,TNF-α,IFN-γ were significantly downregulated(p〈0.05),while those of IL-4 and IL-10 were significantly upregulated(p〈0.05), compared with the model group.The HE staining results showed as follows:the structure of the spinal cord tissues and morphology of nerve cells were intact in the control group; in the model group,abundant inflammatory cells appeared in the spinal cord tissues; in contrast,no obvious inflammatory cells were observed in the BMMSCs group.Taking together,it is suggested that BMMSCs have therapeutic effect on experimental allergic inflammatory response in mice,and the mechanism involves inhibiting the expressions of inflammatory cytokines and reduce the damage to the spinal cord.
