摘 要:目的研究肿瘤坏死因子α诱导蛋白3(tumor necrosis factorα-induced protein 3,TNFAIP3)在胃腺癌组织中的表达及TNFAIP3与胃癌细胞凋亡的关系。方法免疫组织化学检测60例胃腺癌组织和30例正常胃黏膜组织中TNFAIP3表达;Western blot检测两种胃癌细胞株MKN28和SGC7901中TNFAIP3的表达差异,流式细胞术检测两株细胞的凋亡;利用siRNA技术沉默TNFAIP3蛋白的表达后,检测转染后细胞的凋亡。结果 TNFAIP3蛋白在正常胃黏膜组织中多呈低表达,而在胃腺癌组织中多呈高表达。其在低分化腺癌中表达高于高中分化腺癌,而与胃腺癌患者的年龄、性别、浸润深度和淋巴结转移无关。SGC7901细胞中TNFAIP3蛋白表达显著高于MKN28,而SGC7901细胞凋亡率显著低于MKN28细胞。siRNA-TNFAIP3转染SGC7901细胞后,流式细胞术检测显示降低TNFAIP3蛋白表达可显著提高SGC7901的凋亡率。结论胃腺癌组织中高表达TNFAIP3可能通过抑制胃癌的细胞凋亡而参与胃癌的发生和发展。] Objective To explore the expression of TNFAIP3 in gastric adenocarcinoma and the relationship between TNFAIP3 and human gastric carcinoma cell apoptosis. Methods The expression of TNFAIP3 was detected in 60 cases of gastric adenocarcinoma and 30 cases of normal tissue by immunohistochemistry. The expression of TNFAIP3 in two lines of gastric carcinoma cells namely MKN28 and SGC7901was detected by western blot and compared; the apoptosis of the two cell lines was detected by flow cytometry. The expression of TNFAIP3 was then silenced by siRNA technology, and cell apoptosis was detected after transfection. Results The ex-pression of TNFAIP3 was low in most normal gastric mucous tissues, but high in most gastric adenocarcinoma tissues; it was higher in poorly than in highly or moderately differentiated adenocarcinoma, but unrelated to age, gender, depth of invasion and lymphatic me-tastasis; it was much higher in SGC7901 cells than in MKN28 cells, and the apoptosis rate of SGC7901 cells was significantly lower than that of MKN28 cells. Flow cytometry results showed that the apoptosis rate of SGC7901 cells increased significantly with decreased TNFAIP3 expression after siRNA transfection. Conclusions The high expression of TNFAIP3 may be involved in the development of gastric carcinoma through the inhibition of cancer cell apoptosis.
