摘  要:以茄子(Solanum melongena L.)"三月茄"为实验材料,参考NCBI上已公布的SmDFR基因,设计特异性全长引物,克隆SmDFR基因的gDNA全长序列。序列分析结果表明:该基因全长为1 482bp,包含5个外显子和4个内含子,编码区为1 149bp,编码382个氨基酸。蛋白序列相似性分析表明,SmDFR蛋白与马铃薯、辣椒、野生番茄和矮牵牛的DFR蛋白相似性分别为93%、93%、93%和90%。进化树分析表明,SmDFR与甜辣椒、马铃薯、西红柿聚为一类,亲缘关系比较近;与红薯、鱼黄草、田旋花亲缘关系最远。对于研究茄子花青素的生物合成及分子机制具有重要意义,也丰富了不同茄子品种SmDFR基因的相关研究。In this study,we used eggplant'Sanyueqie'(Solanum melongena L.)as materials,and designed a pair of full-length primers based on the published SmDFRgene to clone the integrated genomic DNA(gDNA)of SmDFRgene in'Sanyueqie',and then the sequence of the gDNA was used for bioinformatics analyses.The results showed that the integrated gDNA of SmDFRgene was 1 482 bp,and included five exons and four introns.The full-length CDS of SmDFRgene was 1 149 bp and encoded 382 amino acids.Moreover,the homologies of the DFR protein sequences between potato, pepper, wild tomato, petunia and eggplant'Sanyueqie'were 93%,93%,93% and 90% respectively.The cloning of SmDFRgene from eggplant'Sanyueqie'has an important significance for the research on anthocyanidin biosynthesis and its molecular mechanism in eggplant(Solanum melongena L.),and benefit the research on the correlation of SmDFRgenes derived from different varieties.
