摘  要:目的:研究紫花牡荆素诱导人肝癌HepG2细胞分化作用。方法:体外培养人肝癌Hep G2细胞,采用瑞氏-姬姆萨染色显微镜下测量细胞核质比,放射免疫法检测细胞上清液甲胎蛋白(AFP)水平,酶促反应试剂盒检测细胞碱性磷酸酶(ALP)和γ-谷胺酰转肽酶(γ-GT)的活性;Diamond stone分光光度法测定细胞酪氨酸α-酮戊二酸转氨酶(TAT)的活性。结果:1.0μmol/L紫花牡荆素和全反式维甲酸处理48 h,人肝癌Hep G2细胞核质比降低;AFP的分泌和γ-GT的活性显著降低(P<0.01);ALP和TAT的活性则显著升高(P<0.01)。结论:紫花牡荆素具有诱导人肝癌Hep G2细胞分化作用。Objective To investigate whether casticin induces the differentiation of human hepatocellular carcinoma HepG2 cells.Methods Human hepatocellular carcinoma HepG2 cells were cultured in vitro.The nuclear-cytoplasmic ratio of cells was measured under microscope after Wright-Gimsa staining.The Alpha-fetoprotein(AFP) secretion from cells were assayed by radio immunoassay.The activity of alkaline phosphatase(ALP) and γ-glutamyltranspeptidase(γ-GT) were tested by enzymaticreaction kit.Diamond stone spectrophotometry was employed to determine activity of tyrosine-α-Ketoglutaric acid transaminase(TAT).Results After treatment with 1.0 μmol/L casticin or alltrans tretinoin(ATRA),the nuclear-cytoplasmic ratio of HepG2 cells significantly decreased(P < 0.01).Casticin decreased the secretion of AFP and activity of γ-GT(P < 0.01).In addition,Casticin activated TAT and ALP in Hep G2 cells(P < 0.01).Conclusion Casticin can induce differentiation of human hepatocarcinoma Hep G2 cells.
