摘 要:目的探讨新上市国产肠道病毒71型(enterovirus 71,EV71)灭活疫苗诱导小鼠体液免疫和细胞免疫应答特性。方法分别用我国3家企业生产的EV71灭活疫苗(V1、V2、V3)按0、14 d程序免疫BALB/c小鼠,采用体外微量中和试验检测血清单剂和2剂免疫后14 d的EV71中和抗体效价,ELISA法检测2剂免疫后14 d的小鼠脾单个核细胞(mononuclear cells,MNC)分泌细胞因子水平。结果 3家EV71疫苗1剂免疫后即可诱导90%以上小鼠产生EV71中和抗体阳转,V1疫苗组诱导的中和抗体效价显著高于V2和V3疫苗组,GMT分别为627.1、25.9和75.3(P<0.01);2剂免疫后中和抗体阳转率均为100%,V1和V3疫苗组抗体效价相近,GMT分别为1 752.9和2 013.5(P>0.05),均显著高于V2疫苗组(GMT为228.2,P<0.001)。3家疫苗均可诱导BALB/c小鼠产生Th1和Th2类细胞因子应答,但细胞因子种类具有差异,V1和V3疫苗组诱导小鼠分泌IFNγ的水平显著高于V2疫苗组(P<0.05);3个疫苗组间IL-2分泌水平差异无统计学意义(P>0.05),IL-4和IL-5分泌水平与对照组相比,差异无统计学意义(P>0.05);V2疫苗组诱导小鼠分泌的IL-6水平显著高于V3疫苗组(P<0.01);V3疫苗组诱导小鼠分泌的IL-10水平显著高于V1和V2疫苗组(P<0.05)。结论 3家EV71疫苗均可诱导小鼠产生良好的体液免疫和细胞免疫应答,但呈现出不同的免疫应答特征。Objective To investigate the characteristic of humoral and cellular immune responses of a new licensed inactivated enterovirus 71(EV71) vaccine in mice. Methods BALB/c mice were immunized with inactivated EV71 vaccines V1, V2 and V3 from three manufacturers in China. Each vaccine was inoculated twice on days 0 and 14 respectively. The EV71 neutralizing antibody(Nt Ab)titers of mice on day 14 after the first and second immunization were determined by micro-neutralization test in vitro, while the secretion level of cytokines from mouse spleen mononuclear cells(MNCs)on day 14 after the second immunization by ELISA. Results After the first immunization, the vaccines from three manufacturers induced EV71 Nt Ab in more than 90% of mice. However, the EV71 Nt Ab titer induced by V1(627. 1) was significantly higher than those by V2(25. 9) and V3(75. 3)(P 〈 0. 01). After the second immunization, all the EV71 Nt Ab positive conversion rates in three groups were 100%, while the GMTs induced by V1 and V3 were 1 752. 9 and2 013. 5 respectively(P 〉 0. 05), which were significantly higher than that by V2(228. 2)(P 〈 0. 001). All the vaccines induced the secretions of Th1 and Th2 cytokines in BALB/c mice. However, the types of cytokines were different, and the secretion levels of IFNγ induced by V1 and V3 were significantly higher than that by V2(P 〈0. 05). The IL-2 levels induced by the three vaccines showed no significant difference(P〉 0. 05). The IL-4 and IL-5 levels induced by the vaccines showed no significant difference with that in control group(P 〉0. 05). The IL-6 level induced by V2 was significantly higher than that by V3(P 〈0. 01), while the IL-10 level induced by V3 was significantly higher than those by V1 and V2(P 〈 0. 05). Conclusion All the inactivated EV71 vaccines produced by three manufacturers induced good humoral and cellular immune responses in BALB/c mice. However, the characteristics of responses were different.
