摘 要:目的探讨Klotho对人宫颈癌细胞株Si Ha生物学特性的影响及其作用机制。方法采用RT-PCR及Western blot法检测Klotho基因在宫颈鳞癌细胞中的表达特性。构建Klotho过表达载体,采用CCK-8法、transwell实验和流式细胞仪检测细胞增殖、凋亡和迁移能力的变化;Western blot检测Rho A和ROCK 1的蛋白表达水平。结果 Klotho在宫颈癌细胞系中低表达。在Si Ha细胞中过表达Klotho后,细胞的增殖能力显著降低,周期进程缓慢,而细胞凋亡显著增加;Transwell细胞迁移实验显示,过表达Klotho可抑制Si Ha细胞迁移。此外Western blot实验结果显示,过表达Klotho后,Rho A和ROCK 1的蛋白表达水平均显著下降(P<0.05)。结论 Klotho能够抑制人宫颈癌细胞株Si Ha的增殖和迁移,并促进其凋亡;其作用机制可能与抑制Rho A/ROCK 1信号通路的活化有关,提示Klotho可以作为诊断和靶向治疗宫颈癌的潜在作用位点。Objective To explore the effect of Klotho on biological characteristics of human cervical cancer cell line Si Ha and the relevant mechanisms. Methods RT-PCR and Western blot were used to detect the expression characteristics of Klotho gene in cervical squamous cell carcinoma cells,Klotho overexpression vector was constructed. CCK-8 assay,transwell assay,and flow cytometry were used to detect the changes of cell proliferation,apoptosis,and migration. The expression levels of Rho A protein and ROCK 1 protein were detected by Western blot assay. Results The expression level of Klotho in human cervical cancer cell line Si Ha was low. After overexpression of Klotho in Si Ha cells,cell proliferation ability reduced,cell cycle was slow,and cell apoptosis increased significantly. Transwell assay showed that overexpression of Klotho inhibited migration of Si Ha cells. Western blot assay showed that the expression levels of Rho A protein and ROCK 1 protein decreased significantly after overexpression of Klotho( P〈0. 05). Conclusion Klotho can inhibit migration and metastasis of human cervical cancer cell line Si Ha and promote cell apoptosis. The mechanism may be correlated with activation of Rho A/ROCK 1 signal pathway,which indicates that Klotho can be used as a potential new target for diagnosis and targeted therapy of cervical squamous cell carcinoma.
