幽门螺杆菌感染者IL-8-251多态性及胃液相关细胞因子分泌

摘　　要：目的探讨幽门螺杆菌感染者IL-8-251各基因型与胃液中血管生成相关的细胞因子分泌之间的相关性。方法摘取正常人和胃癌患者胃黏膜组织以PCR-RFLP技术对IL-8-251进行基因分型;ELISA法检测胃分泌液中IL-8、VEGF和MMP-9细胞因子浓度。结果 HP(-)正常对照组中,AA型:IL-8(14.4±1.2)μmol/l,VEGF(2.31±0.48)μmol/l,MMP-9(20.4±5.3)μmol/l;TT型:IL-8(8.7±2.6)μmol/l,VEGF(1.28±0.33)μmol/l,MMP-9(8.9±3.5)μmol/l。HP(+)对照组:AA型:IL-8(22.6±5.3)μmol/l,VEGF(3.22±0.62)μmol/l,MMP-9(27.2±6.7)μmol/l;TT型:IL-8(14.2±3.6)μmol/l,VEGF(1.44±0.38)μmol/l,MMP-9(10.6±4.4)μmol/l。HP(-)胃癌组:AA型:IL-8(32.8±6.6)μmol/l,VEGF(3.81±0.72)μmol/l,MMP-9(46.2±8.3)μmol/l;TT型:IL-8(22.3±4.5)μmol/l,VEGF(1.83±0.42)μmol/l,MMP(20.2±4.6)μmol/l。HP(+)胃癌组:AA型:IL-8(38.2±5.6)μmol/l,VEGF(4.62±0.76)μmol/l,MMP-9(62.6±13.2)μmol/l;TT型:IL-8(28.6±3.8)μmol/l,VEGF(2.54±0.45)μmol/l,MMP-9(32.1±6.8)μmol/l。结论伴随HP感染者IL-8-251 AA型胃液中血管生成相关细胞因子IL-8、VEGF和MMP-9分泌增强,提示IL-8-251 AA型且HP感染者可能更易促使胃癌的发生发展。Objective：To investigate the correlation between IL-8-251 genotypes and the secretion of cytokines related to blood vessels in gastric juice. Methods ：The gastric mucosal tissue was obtained with endoscopy and the genotype of IL-8- 251 and analyzed with PCR-RFLP; also the concentration of IL-8, VEGF and MMP-9 in gastric juice were detected with ELISA. HP （ - ） gastric cancer group： type AA： IL-8 （32. 8 ± 6. 6 ） p, mol/1, VEGF （ 3.81 ± 0. 72 ） μmol/1 MMP-9 （46. 2 ± 8. 3 ） μmol/1; TT type： IL-8 （22. 3 ±4. 5） μmol/1, VEGF （ 1.83 ±0. 42） μmol/1, MMP （20. 2 ±4. 6） μmol/l. HP （ ± ） group of gastric cancer：AA type： IL-8 （38. 2 ± 5.. 6） μ mol/1, VEGF （4. 62 ± 0. 76） μmol/1, MMP-9 （62, 6 ± 13.2）μmol/1 ; TT type： IL-8 （28. 6 ± 3.8） μmol/l, VEGF（2. 54 ± 0. 45 ） μmol/1, MMP-9 （32. 1 ± 6.8 ） μmol/l. Conclusion： The cytokines related to blood vessels（ IL-8,VEGF and MMP-9）in gastric juice secretion of IL-8-251 type AA group with HP infection increases, which suggest that AA type with HP infection may occur more likely to induce gastric cancer.
