摘　　要：目的:探讨沉默MSP58基因对肺癌细胞A549增值,细胞周期和侵袭能力的影响。方法:化学合成针对MSP58基因的siRNA,转染肺癌细胞A549,RT-PCR和Western-blot检测MSP58基因的mRNA和蛋白水平,利用MTT法绘制细胞生长曲线,流式细胞仪分析细胞周期,Transwell小室实验观察侵袭能力。结果:针对MSP58基因的siRNA转染A549细胞48h后,与阴性对照组和空白对照组相比,siRNA组显著降低MSP58的基因表达和蛋白表达,细胞的增值受到抑制,并随着时间延长抑制明显,细胞发生G1期阻滞,细胞的侵袭力下降。结论:利用特异性siRNA沉默MSP58基因的表达,可以显著抑制肺癌A549细胞增值和侵袭能力,MSP58有可能成为肺癌治疗的新靶点。Objective：To determine the effect of MSP58 knockdown by RNA interference （RNAi） on the proliferation and migration in lung cancer cells A549.Methods：The siRNA targeting MSP58 gene and negative control siRNA were synthesized and transfected to lung cancer cells A549.RT-PCR and Western-blot were applied to observe the expression of mRNA and protein level of MSP58.The cell growth curve was drawn by MTT.Cell cycle was analyzed by flow cytometry.The cell migration ability was detected by Transwell assay.Results： After transfected 48h, comparing to the negative group and blank group, the expression of MSP58 mRNA and protein was down-regulated by siRNA targeting MSP58 gene.The cell proliferation of A549 in siRNA group was suppressed in a time-dependent manner, cell cycle was blocked in G1 phrase, and the ability of migration declined significantly.Conclusion： Specific siRNA targeting MSP58 efficiently inhibits the proliferation and migration in A549 cells,and the MSP58 gene is a promising anti-tumor target of lung cancer.
