用高转移人骨肉瘤亚群OS-732建立裸鼠骨肉瘤肺转移模型

摘　　要：目的建立一种发病过程类似人类骨肉瘤肺转移的动物模型。方法分别采用5×10~5、1×10~6、5×10~6三个剂量的高转移人骨肉瘤亚群OS-732接种裸鼠皮下,饲养8周。测量局部肿瘤和肺转移结节。结果 5×10~5组无论是局部成瘤率和肺转移率均为0。1×10~6组和5×10~6组局部成瘤率均为100%,肺转移率均为87.5%。从后二者的生长曲线上看,局部肿瘤的生长趋势大致相同,在第8周时,两组局部肿瘤的湿重和平均直径无显著差异(P>0.05)。可以认为两组的局部肿瘤生长趋势无显著差异。从两组的肺转移情况看,两组的肺转移率均为87.5%,肺表面结节数无显著性差异,肺结节总转移数无显著性差异(P>0.05)。结论 1×10~6、5×10~6剂量的高转移人骨肉瘤亚群OS-732接种裸鼠皮下均可建成满意的骨肉瘤肺转移模型。从动物生存时间和转移的专一性上看,1×10~6OS-732的剂量更加合理,建立的模型更为实用,是研究骨肉瘤肺转移理想的动物模型。Objective To establish an animal model of osteosarcoma with lung metastasis. Methods Incubate the OS -7 3 2 cell line on the nude mice subcutaneously with the amount 5 ×10^5 , 1 ×10^6 and 5 ×10° respectively. Observe the mean diameters, wet weight of the primary tumors. At the same time, the lung metastasis nodules and calculate the total metastasis nodules were observed. Results The incidences of 1 ×10^6 group and 5 ×10^6 group are 100% , lung metastatic rates are 87.5%. there are no significant differences of the mean diameters, wet weight of the primary tumors and lung metastasis nodules between 1 ×10^6 group and 5 X 106 group（ P 〉0. 05） . Conclusion The nude mice model of the dosages of 1 ×10^6 and 5 ×10^6 of OS - 732 cell are satisfied and the former is more reasonable. The nude mice model with the dosage may provide particular advantage for investigating treatments of the osteosarcoma lung metastasis.
