摘  要:目的罗勒多糖具有抗肿瘤侵袭转移、化疗增效以及降血糖、降血脂和抗血栓等多种生物活性作用。本研究探讨罗勒多糖对转化生长因子-β(transforming growth factor-beta,TGF-β)诱导的人肺癌A549细胞侵袭运动能力的抑制作用,揭示罗勒多糖抑制上皮间质转化作用的可能机制。方法体外培养肺癌A549细胞,实验分为3组:(1)正常对照组,不加任何处理;(2)模型组,TGF-β5ng/mL;(3)罗勒多糖组,TGF-β5ng/mL+罗勒多糖200μg/mL。免疫荧光检测罗勒多糖对E-cadherin、Vimentin和α-SMA表达的影响,细胞侵袭实验和运动实验检测细胞对人工基底膜的降解和运动能力,细胞划痕实验检测细胞运动能力。结果与正常对照组比较,模型组细胞Vimentin和α-SMA表达增强,E-cadherin表达减弱。罗勒多糖处理后Vimentin和α-SMA表达减弱,E-cadherin表达增强。TGF-β可提高细胞降解人工基底膜的能力,单位视野下侵袭至下室的细胞数模型组为69.0±4.3,与正常对照组(42.0±4.30)和罗勒多糖组(37.0±4.41)比较,差异有统计学意义,均P<0.05。TGF-β增强了细胞的侵袭能力,罗勒多糖可以抑制TGF-β诱导的A549细胞侵袭能力。TGF-β使FN趋化的迁移细胞数目增多,单位视野下侵袭至下室的细胞数模型组为95.2±3.83,与正常对照组(69.2±5.11)和罗勒多糖组(68.2±6.18)比较,差异均有统计学意义,均P<0.05。划痕实验显示,TGF-β增强了细胞的迁移能力,罗勒多糖可以抑制TGF-β诱导的A549细胞迁移能力。结论罗勒多糖能够调节A549细胞上皮间质转化,这可能是其抑制A549细胞的侵袭转移能力的重要机制。OBJECTIVE Basil polysaccharide has many bioactivities, including anti-tumor invasion and metastasis, synergistic effect of chemotherapy,reducing blood sugar and blood fat as well as anti thrombus. The objective of this study was to study the basil polysaccharide inhibited transforming growth factor-beta(TGF-13) induced A549 cell invasion and motility and study the mechanism of basil polysaccharide inhibited epithelial mesenchymal transition function. METHONDS Effect of basil polysaccharide on the immune fluorescence detection of E-cadherin, Vimentin, ccSMA expression, cell invasion test and exercise test were used to detect the degrada- tion and movement of cells on the artificial basement membrane, cell scratch assay was used to detect cell motility. RESULTS Com- pared with the normal control group,the expression of Vimentin and a-SMA in the model group increased while that of E-cadherin decreased. In the cells treated with basil polysaccharide,the up-regulation of Vimentin as well as a-SMA expression and down-regula- tion of the E-cadherin expression induced by TGF--β was inhibited. TGF-β could promote the cells' ability of degrading artificial base- ment membrane. In the model group,the average number of cells invaded to the lower chamber in one visual field under microscope was 69.0±4. 3, and that in the normal control group was 42.0±4. 30 ,which showed that the difference was statistically significant (P〈0. 05). Basil polysaccharide completely repressed the TGF-β induced cell invasion, the average number of cells on the lower membrane was 37. 0±4. 41. Cell migration assay showed that average number of cells under the vision of migrated to the lower sur- face of chamber membrane was 95.2±3.83 in the model group,69.2±5. 11 in the normal control group and 68. 2±6. 18 in the basil polysaccharide group,there was statistically significance between the normal control group and the model group (P〈0. 05) or thebasil polysaccharide group (P〈0.05) and the model group.
