摘  要:肝星状细胞(HSCs)又称储脂细胞、Ito细胞,是肝脏固有非实质细胞类型之一,在肝脏细胞中所占比例不到8%。它存在于肝窦内皮细胞与肝细胞之间的Disse间隙内,并以脂滴的形式储存人体维生素A。HSCs是细胞外基质的主要来源,它的活化是肝纤维化发生发展过程中的关键环节。原代HSCs分离方法的研究是进行肝纤维化发生机制研究的基础,是肝纤维化研究的热点之一。目前关于HSCs的分离方法有经典的肝脏二步酶灌注法、流式细胞术分选法、磁珠分选法等,本文就近年来原代HSCs分离方法新进展进行综述。Hepatic stellate cells ( HSCs), also called fat-storing cells or Ito cells, are one of inherent liver non-parenchymal cell types. They constitute 〈 8% of the total liver cell population. They locate in the Disse space between liver sinusoidal endothelial ceils and hepatocytes, and store Vitamin A in the form of lipid drops. HSCs are the main source of extracellular matrix. Their activation is the key process in the development of liver fibrosis. The isolation of primary HSCs is the basis of studying the mechanism of liver fibrosis. It is one of the research focus on the area of liver fibrosis. The methods of primary HSCs isolation include the classic two-step enzyme perfusion,fluorescence activated cell sorting and magnetic-activated cell- sorting. This review will summarize the new progresses of primary HSCs isolation in recent years.
