四妙勇安汤抑制血管内皮细胞P-选择素、NF-κB表达的实验研究

摘　　要：目的:探讨四妙勇安汤及其单体绿原酸、阿魏酸对于凝血酶处理的内皮细胞P-选择素及NF-κb p65mRNA表达的影响。方法:MTT法检测绿原酸、阿魏酸、四妙勇安汤对于内皮细胞活力的影响;0.2 U/mL凝血酶处理内皮细胞15 min作为模型组,3种药物预处理内皮细胞24 h作为干预组,流式细胞术、Elisa检测内皮细胞P-选择素的表达,RT-PCR检测NF-κb p65mRNA的表达。结果:绿原酸102 ng/mL、阿魏酸104 ng/mL时OD值最大;模型组P-选择素及NF-κb p65mRNA的表达均高于空白组(P<0.05),各干预组P-选择素及NF-κb p65mRNA的表达均低于模型组,组间无差异,但四妙组数值低于其余两组。结论:四妙勇安汤及其单体能减少内皮细胞P-选择素的表达,这一作用可能是通过抑制NF-κb p65mRNA的表达来完成的。Objective：To investigate the effect of Simiao Yong’an Deoction and its monomer green chlorogenic acid and ferulic acid on the expression of the thrombin treated endothelial cell P-selectin and NF-κb p65 mRNA.Methods：MTT assay tested the influence of green chlorogenic acid,ferulic acid and Simiao Yong’an Deoction on endothelial cell viability.The endothelial cells treated with 0.2 U/mL thrombin for 15 min were as a model group.The endothelial cells pretreated by these three drugs for 24 h were as the intervention group.The flow cytometry and ELISA detected endothelial cell P-selectin expression and RT-PCR detected the expression of NF-κb p65 mRNA.Results：When chlorogenic acid was 102 ng/mL and the ferulic acid 104 ng/mL,the OD value was the maximum.The model group’s P-selectin and NF-κb p65 mRNA expression were higher than those of the blank group（P〈0.05）.Each intervention group’s P-selectin and NF-κb p65 mRNA expression were lower than those of the model group,and there were no differences between groups.But the value in the Simiao Yong’an Deoction was lower than the other two groups.Conclusion：Simiao Yong’an Deoction and its monomer can reduce P-selectin expression in endothelial cells,which might be accomplished by inhibiting the expression of NF-κb p65 mRNA.
