摘 要:目的:观察肉桂醛对高脂饮食C57BL/6小鼠骨代谢的影响,探讨其与组织蛋白酶K(Cathepsin K)表达的相关性。方法:肉桂醛治疗高脂喂养的C57BL/6小鼠8周后,分别取小鼠的股骨和胫骨,用HE染色和茜素红染色观察骨微结构变化、骨代谢以及骨发育情况;用质构仪评价骨生物力学性能;用免疫组织化学法观察Cathepsin K在骨组织中的表达情况。结果:肉桂醛治疗8周后,高脂饮食小鼠骨小梁断裂程度减轻,排列整齐,骨形成明显增多(P<0.05);骨的硬度形变量、第一循环硬度和峰值压力均明显升高。同时,肉桂醛能明显降低骨组织Cathepsin K蛋白表达(P<0.05)。结论:肉桂醛能改善高脂饮食喂养的C57BL/6小鼠骨强度,改善骨微结构,改善骨代谢,其作用机制可能是与调节Cathepsin K的表达有关。Objective:To observe the effect of cinnamaldehyde on bone quality changes in high fat diet fed C57BL/6mice,so as to investigate the correlation of cinnamaldehyde and expression of Cathepsin K.Methods:C57BL/6 mice were fed with high fat diet.After 8 weeks of treatment with cinnamaldehyde,HE staining and alizarin red staining(Alizarin Red S Stain) were used to observe the bone microstructure changes,bone metabolism and bone growth of femur and tibia in mice.Texture analyzer was used to evaluate biomechanical properties.By immunohistochemical staining,the expression of Cathepsin K protein was observed.Results:After 8 weeks of treatment with cinnamaldehyde,the fracture degree of the trabecular bone was reduced and the trabecular bone was arranged regularly in cinnamaldehyde group.Furthermore,bone formation significantly increased(P〈0.05).Hardness dcformation,the first cycle degree of hardness and peak pressure of the femur in mice were also significantly increased(P〈0.05).At the same time,the expression of Cathepsin K in bone tissue was significantly decreased in cinnamaldehyde group(P〈0.05).Conclusion:Cinnamaldehyde can improve bone strength,bone microstructure and bone metabolism in high fat induced C57BL/6 mice.The mechanism of action may be related to the regulation of Cathepsin K expression.
