摘 要:为探讨灵芝多糖硫酸酯(GLPS)对D-半乳糖致衰小鼠学习记忆能力的影响,本试验对灵芝多糖(GLP)进行硫酸酯化修饰,将50只昆明系小鼠随机分为5组:空白对照组、模型对照组、阳性对照组(吡拉西坦片50 mg/kg体重·d)、GLP给药组(50 mg/kg体重·d)、GLPS给药组(50 mg/kg体重·d),建立D-半乳糖致衰小鼠模型。进行跳台试验,并检测小鼠脑组织SOD活性和MDA含量的变化;免疫印迹检测小鼠海马组织Bcl-2和Caspase-3的影响。结果发现,GLP和GLPS组均能够明显延长小鼠跳台潜伏期,减少5 min内出现的错误次数,且均可升高SOD活性、降低MDA含量,但GLPS组效果明显优于GLP组;免疫印迹结果表明,GLPS组能够明显降低Caspase-3的表达,增强Bcl-2蛋白的表达,而GLP组影响不明显。说明GLPS可以显著改善D-半乳糖致衰小鼠的学习记忆能力,其机制可能与增强Bcl-2的表达及抑制Caspase-3的表达,抑制海马组织凋亡相关。To study the effect of GLPS on the learning and memory abilities of D-galactose-induced senile mice, GLP was modified by sulfation to obtain GLPS. Fifty KM mice were randomly divided into 5 groups: blank control group, model control group, positive control group( laci staw 50mg/kg · d).,GLP group(50 mg/kg · d) and GLPS group (50 mg/kg, d). We established the D-ga- lactose-induced senile mice model and performed the step-down passive avoidance test. And all mice were beheaded to detect the effect of SOD and MDA. The expression of easpase-3 and Bcl-2 in the hippocampus of mice was detected by Western blotting. The results showed the GLP and GLPS group increased the incubation period platform, and decreased the mistaken reaction times in 5 min. We also found that GLP and GLPs group increased SOD activity, and decreased MDA content, but the effect of GLPS group was better than GLP group. The result of Western blotting showed that the GLPS group significantly reduced the expression of caspase-3, and increased the expression of Bcl-2 protein. These results showed that sulfation can significantly improve the effect of GLP on the learning and memory abilities of D-galactose-induced senile mice. The possible mechanism is that GLPS can depress the oxidative stress and inflammatory response induced by scopolamine through the affection of NF-kB signal transduction pathway.
