摘 要:目的检测PTEN、Bcl-2及Cyclin D1在口腔鳞癌中的表达,并将PTEN蛋白表达与Bcl-2及Cyclin D1的蛋白表达相比较,探讨PTEN/PI3K/AKT信号通路在口腔鳞癌发生发展中的分子机制。方法采用免疫组化,检测32例口腔鳞癌和4例正常口腔黏膜组织中PTEN、Bcl-2及Cyclin D1蛋白的表达水平,将PTEN蛋白表达与临床病理参数进行相关分析,并分析PTEN与Bcl-2及Cyclin D1蛋白表达的相关性。结果口腔鳞癌中PTEN蛋白的表达率为46.9%(15/32);口腔鳞癌中Bcl-2蛋白的阳性率为53.1%(17/32);口腔鳞癌中Cyclin D1蛋白的阳性率为50.0%(16/32)。PTEN蛋白的缺失表达与细胞分化程度密切相关。PTEN缺失表达的患者绝大多数是中低分化的口腔鳞癌,中低分化的口腔鳞癌的PTEN表达水平比高分化者明显降低(P<0.05)。口腔鳞癌中PTEN蛋白的表达与Cyclin D1,Bcl-2蛋白表达均呈负相关关系(P<0.05)。结论PTEN,Cyclin D1,Bcl-2在口腔鳞癌的发生过程中起着重要的作用并相互之间有密切的关系,可能作为分子靶向治疗的靶点。Objective To investigate the macular machinism of PTEN/PI3K/AKT pathway in the generating process of OSCC by the relationship among the expression of PEN, Bel-2, CyclinD1 protein. Methods The immunohistochemical stains against PTEN, Bcl-2, and CyclinD1 were performed to determine expression of protein in 32 cases of OSCC and 4 cases of normal oral mucosa. All results were analyzed quantitatively by imagine analysis technique. Then, the results were compared with clinicopathological parameters. Results The positive expression of PTEN protein in normal oral mueosa and OSCC was 100% and 46.9%. The positive expression of Bcl-2 protein in normal oral mucosa and OSCC was 0% and 53.1%.The positive expression of CyclinD1 protein in normal oral mueosa and OSCC was 0% and 50.0%. The expression of PTEN of OSCC in middle differential and low differential OSCC was significantly lower than that of good differential OSCC (P〈0.05). There was significant negative correlation between the expression of PTEN and the expression of Bcl-2, CyclinD1. Conclusion The low expression of PTEN in OSCC could predict a poor prognosis. PTEN, Bcl-2, and CyclinD1 maybe paly an impotant role in the generating process of OSCC and could be used as a marker for molecular targeting treatment.
