摘 要:目的:建立T细胞免疫功能重建模型,研究免疫与胰腺癌淋巴转移的关系,探讨免疫的影响因素。方法 :建立裸鼠胰腺癌淋巴转移模型及T细胞免疫功能重建的模型。将裸鼠分为免疫功能重建组(R1)和免疫功能未重建组(R2)。比较两组成瘤时间差异。病理切片观察淋巴结转移程度。流式细胞仪检测两组裸鼠外周血及脾脏CD3+CD4+、CD3+CD8+细胞百分比。结果 :R1组成瘤时间(8.5±0.6)d,R2组成瘤时间(6.4±0.8)d,两者差异具有统计学意义(P<0.05)。R1组胰腺癌淋巴转移受到一定程度的抑制,髂总动脉旁淋巴结的转移率明显低于R2组(P<0.05)。R1组裸鼠外周血以及脾脏CD3+CD4+、CD3+CD8+细胞百分比明显高于R2组。结论:T细胞免疫功能重建裸鼠模型较好地模拟免疫环境,为胰腺癌淋巴转移的免疫相关研究提供动物实验平台。Objective To provide T lymphocyte model of immunological function reconstruction and to study the association of immunity with lymphatic metastasis of pancreatic cancer. Methods Both the nude mice model of pancreatic cancer with lymphatic metastasis and T lymphocyte model of immunological functions reconstruction were established.Nude mice were divided into 2 groups. The immunological function was reconstructed in the nude mice as R1 group and the nude mice without immunological functions reconstruction were as R2 group. The time of tumor formation in the nude mice were compared between 2 groups. Lymph nodes metastases in nude mice were examined histologically. CD3+CD4+and CD3+CD8+ in the peripheral blood and spleen of nude mice were detected by flow cytometry. Results The time of tumor formation was(8.5±0.6) days in R1 group and(6.4±0.8) days in R2 group, and the difference was statistically significant(P〈0.05). Lymphatic metastases of pancreatic cancer were inhibited at some extent in the nude mice in R1 group.The rate of metastases to common iliac artery lymph nodes in R1 group was higher than that in R2 group. The ratio of CD3+CD4+ and CD3+CD8+ in the peripheral blood and spleen of the nude mice in R1 group was higher than that in R2 group significantly. Conclusions The model of T lymphocyte in nude mice with immunological functions reconstruction in this study could provide a good immunological platform for the study on immunological functions associated with lymphatic metastasis of pancreatic cancer.
