摘 要:目的建立半夏组培快繁体系。方法以半夏茎尖、叶柄和叶片为外植体,研究不同消毒方法和激素组合对半夏组织培养的影响。结果以75%酒精30s+(2%次氯酸钠+1.67g/L头孢拉定)15min处理外植体为宜;茎尖有利于不定芽的诱导;不定芽诱导最佳培养基为MS+1 mg/L 6-BA+0.3 mg/L NAA+0.6 mg/L KT;高浓度6-BA和低浓度NAA有助于半夏不定芽和块茎的形成;生根培养以1/2MS+0.5 mg/L IBA为最佳。结论建立了半夏组培快繁体系,为利用生物反应器生产半夏小块茎提供了理论依据。Objective To establish the system of tissue culture and rapid propagation for Pinellia ternata. Methods Taking stem tip,petiole and leaf of Pinellia ternata as explants to study the effect of different disinfection methods and hormone combinations on tissue culture of Pinellia ternata. Results Explants were treated with 75% ethanol for 30 s + 2% sodium hypochlorite and 1.67g/L Cefradine for 15 min. Stem tips were beneficial to induce adventitious buds. The optimal medium for induction of adventitious buds was MS + 1 mg/L 6-BA + 0. 3 mg/L NAA + 0. 6 mg/L KT. High-concentration 6-BA and low-concentration NAA were conducive to adventitious bud and tuber formation of Pinellia ternata. 1/2MS + 0. 5 mg/L IBA was suitable for rooting culture. Conclusion The system of tissue culture and rapid propagation of Pinellia ternata was established,which provided a theoretical basis for the production of Pinellia ternata tuber with bioreactors.
